Feasibility study of Cooral system for oral mucositis in patients receiving chemotherapy

ABSTRACT: Interventions: Before the start of treatment the patient receives clear oral and written instructions on the use of Cooral by the nurse responsible for the patient. The patient him/herself is able to administer the intraoral component until it feels comfortable. Then the responsible staff check to ensure that it has good contact with the oral mucous membrane. Cooling begins 30 minutes before the start of chemotherapy. Cooling continues until 30 minutes after the termination of the cytostatic infusion. During treatment the patient may if necessary take out the component and replace it again, for a maximum of 10 minutes. Food should thus be taken before or after chemotherapy. Primary outcome(s): Feasibility for Cooral system during the first chemotherapy cycle. Study Design: Single arm Non-randomized

RNA-seq characterization of an infection of Mycobacterium smegmatis by a cluster A4 mycobacteriophage

Project description:We present transcriptomic analysis of an infection by a cluster A4 mycobacteriophage of its host, Mycobacterium smegmatis, at six time points (n = 2). Viral transcription is detectable 5 minutes into infection, and viral mRNA makes up the vast majority of all transcription by 30 minutes. Kampy, like many phages, has a genome organized into two arms bearing early- and late-phase genes. We identify two transcriptional start sites (TSS’s) consistent with an early promoter driving expression of half of the genome, along with a second, late-stage promoter that becomes active around 30 minutes after the start of infection.

The transcriptome profiling and analysis of CAFs isolated from breast cancer skin metastasis, cutaneous basal cell carcinoma and squamous cell carcinoma arising from oral cavity mucous membrane unravelled major gene candidates such as the IL-6, VEGF-A and MFGE8 that induced the expression of several markers associated with poor prognosis and epithelial-to-mesenchymal transition in co-cultivated EM-G3 breast cancer cells.

Project description:Patient selection and specimen collection. Thirty-six freshly frozen tumor samples were prospectively collected from patients undergoing surgery or biopsy for HNSCC at the University of North Carolina (UNC) at Chapel Hill (21 patients) and Vanderbilt University (15 patients). All tissues were snap-frozen in liquid nitrogen within 30 minutes of surgical resection or biopsy, and kept at -80oC until further processing. All patients consented to participation in this study under protocols approved by IRB at the two institutions. This set includes the samples previously deposited into GEO including 8 additional samples and adjustment for protocol changes. Attenuated inflammatory profile following single and repeated handgrip exercise and remote ischemic preconditioning in patients with cerebral small vessel disease

Project description:Similar to remote ischemic preconditioning bouts of exercise may possess immediate protective effects against ischemia-reperfusion injury. However, underlying mechanisms are largely unknown. This study compared the impact of single and repeated handgrip exercise versus remote ischemic preconditioning on inflammatory biomarkers in patients with cerebral small vessel disease (cSVD). In this crossover study, 14 patients with cSVD were included. All participants performed 4-days of handgrip exercise (4x5-minutes at 30% of maximal handgrip strength) and remote ischemic preconditioning (rIPC; 4x5-minutes cuff occlusion around the upper arm) twice daily. Patients were randomized to start with either handgrip exercise or rIPC and the two interventions were separated by >9 days. Venous blood was drawn before and after one intervention, and after 4-days of repeated exposure. We performed a targeted proteomics on inflammation markers in all blood samples.

Project description:We performed a comprehensive study on 10 extensively studied mine rescue team members before and after a 30 minutes stay in the high temperature simulated environment, utilizing longitudinal multi-omic profiling. To carry out the multi-omics analysis, intravenous blood samples were collected at various time points: before exposing the members to acute heat stress, as well as at 5 minutes, 30 minutes, 1 hour, and 24 hours during the recovery process. In-depth proteomics profiling was performed on each sample.

Insulin mitigates acute ischemia induced atrial fibrillation and sinoatrial node dysfunction ex vivo

Project description:Acute atrial ischemia is a well-known cause of postoperative atrial fibrillation (POAF). However, the cellular and electrophysiological mechanisms through which ischemia contributes to the development of POAF are not well understood. Administering insulin 30 minutes before ischemia and during reperfusion with 25 minutes of ischemia greatly reduced the vulnerability to AF.
