Project description:IL-4 and IL-13 are cytokines involved in type 2 immune responses involved in atopic asthma and other diseases. They have a partially overlapping set of cognate receptors whose roles are incompletely understood. Here we explore the effects of blocking one or both subunits of the IL-13 alpha receptor on the action of these cytokines on IMR-90 lung fibroblast cells. IMR-90 lung fibroblast cells were cultured on matrigel and treated with various combinations of IL-4 (10 ng/ml), IL-13 (10ng/ml), 228 B/C (which blocks signaling from IL13RA1), and 11H4 (which blocks signaling from IL13RA1 and IL13RA2).

Project description:Chlorella has been used as a food supplement and has been reported to have health benefits. We used the forced swimming test to investigate the influence of Chlorella intake during muscle stress training in mice. Swimming time was about two-fold longer for the Chlorella intake mice than for the control mice. Microarray analysis revealed that the global expression profile of muscle from the Chlorella intake mice was similar to that for intact (nonswimming) mice rather than to that for control swimming mice. By contrast, oxidoreductase activity and the leukotriene synthesis pathway were repressed in Chlorella intake mice. According to measurement of the cytokine activities of primary cultures of spleen, Th1 cytokines, such as interferon-gM-cM-^CM-;and interleukin-2, and granulocyte colony-stimulating factors, such as MIP-1aM-cM-^CM-; GMCSF, and IP-10, were decreased in control swimming mice, but these cytokines were rescued in the Chlorella intake mice. We suggest that the immune response during muscle training can be repressed by intake of Chlorella. The intact (nonswimming) and control (swimming) groups were fed normal food, and the Chlorella intake (swimming) group was fed food including 0.5% Chlorella powder. After 14days, the total RNA was isolated from muscle and samples was pooled for each group (n = 10 for each group).

Project description:We report a fully human UC-derived organoid system that integrates ECM hydrogel, MSCs and reprogrammed hepatocytes, achieving the treatment of acute hepatic failure in case of extreme failure.To explore the mechanism of our organoid in hepatic failure, we performed single cell RNA sequencing of transplanted organoids as well as the original organoid prior to transplantation, we have demonstrated that MSCs were primarily responsible for inhibiting the secretion of inflammatory-inducing cytokines and promoting the secretion of inflammation-inhibiting cytokines by recruiting immune cells migrating into organoid to triggering immune response.

Project description:Dominant theories of depression position self-concept as a central determinant of psychological functioning, but the relationship between the structure of self-concept and depression has not been extensively explored. The present study investigates the relationship between the structure of the self and psychopathological outcomes (depressive symptoms and neuroticism) with two methodological approaches. Using an established framework that draws insight from Buddhist psychology, the structure of the self is conceptualized in terms of selflessness and self-centeredness. Specifically, selflessness is construed as a multidimensional concept characterized by interdependence, outsider phenomenology, and impermanence. The three dimensions of the self were assessed at age 26 with inventories of self-transcendence, perspective-taking, and materialism, respectively (N = 814). First, a variable-centered approach was used to investigate potential interactions between the dimensions of selflessness. Self-transcendence negatively predicted depressive symptoms and neuroticism, whereas perspective-taking and materialism were positively associated with the outcomes. Self-transcendence moderated the relationship between perspective-taking and depressive symptoms. Perspective-taking was not statistically related to depressive symptoms for participants who exhibited higher levels of self-transcendence. The results clarify ambiguous associations between perspective-taking and depression found in previous research. Second, person-centered analyses were used to identify five profiles of self-structure: (1) Selfless, (2) Selfless Materialist, (3) Interdependent Insider, (4) Self-centered Non-materialist, and (5) Self-centered. As hypothesized, the Selfless cluster was associated with low levels of depressive symptoms and neuroticism, whereas the Self-centered cluster was associated with high levels. The profiles demonstrate the manifestation of several combinations of features of the self, which contributes to overall understanding of selflessness by complicating the traditional dichotomy between selflessness and self-centeredness.

Project description:Background: The immunologic response to stress is regulated by the cytokines. Anti-Tumor Necrosis Factor-α agents are antibodies directed against a key cytokine in the process angiogenesis and collagen synthesis. It is not known whether they intervene with surgical stress response increasing the rate of postoperative complications.
Method: Un-blinded prospective, non-interventional cohort single centre study including all the patients with Crohn’s disease and Ulcerative Colitis undergoing abdominal surgery. Immunological and endocrinological parameters will measured in blood samples taken from these patients before and after surgery. Power calculations showed that 17 patients in each arm are needed.

Project description:Objectives: Pinolenic acid (PNLA), an omega-6 polyunsaturated fatty acid from pine nuts, has anti-inflammatory and anti-atherogenic effects. We aimed to investigate the direct anti-inflammatory effect and anti-atherogenic effects of PNLA on activated purified CD14 monocytes from peripheral blood of patients with rheumatoid arthritis (RA) in vitro. Methods: Flow cytometry was used to assess the proportions of CD14 monocytes expressing TNF-α, IL-6, IL-1β, and IL-8 in purified monocytes from patients with RA after lipopolysaccharide (LPS) stimulation with/without PNLA pre-treatment. The whole genomic transcriptome (WGT) profile of PNLA-treated, and LPS-activated monocytes from patients with active RA was investigated by RNA-sequencing. Results: PNLA reduced percentage of monocytes expressing cytokines: TNF-a by 23% (p=0.048), IL-6 by 25% (p=0.011), IL-1B by 23% (p=0.050), IL-8 by 20% (p=0.066). Pathway analysis identified upstream activation of peroxisomes proliferator-activated receptors (PPARs), sirtuin3, and let7miRNA, KLF15 which are anti-inflammatory and antioxidative. In contrast, DAP3, LIF and STAT3, which are involved in TNF-a, and IL-6 signal transduction, were inhibited. Canonical Pathway analysis showed that PNLA inhibited oxidative phosphorylation (p=9.14E-09) and mitochondrial dysfunction (p=4.18E-08), while the sirtuin (SIRTs) signalling pathway was activated (p=8.89E-06) which interfere with the pathophysiologic process of atherosclerosis. Many miRNAs were modulated by PNLA suggesting potential post-transcriptional regulation of metabolic and immune response that has not been described previously. Multiple miRNAs target pyruvate dehydrogenase kinase-4 (PDK4), single-immunoglobulin interleukin-1 receptor molecule (SIGIRR), mitochondrially encoded ATP synthase membrane subunit 6 (MT-ATP6) and Acetyl-CoA Acyltranferase2 (ACAA2); genes implicated in regulation of lipid and cell metabolism, inflammation, and mitochondrial dysfunction. Conclusion: PNLA has potential anti-atherogenic and immune-metabolic effects on monocytes that are pathogenic in RA and atherosclerosis. Dietary PNLA supplementation regulates key miRNAs that are involved in metabolic, mitochondrial, and inflammatory pathways.

Project description:Not all depression patients effectively respond to repeated transcranial magnetic stimulation (rTMS). We tested whether the intrinsic functional connectivity (FC) strength between the stimulated left dorsolateral prefrontal cortex (DLPFC) and left nucleus accumbens (NAcc) might predict effects of rTMS. Twenty-two medication-naïve depression patients received rTMS on left DLPFC for 2 weeks and underwent baseline functional magnetic resonance imaging (fMRI). We compared the amplitude of the low-frequency fluctuation (ALFF) and regional homogeneity (ReHo) in the stimulated target (the cortex region directly stimulated by rTMS) located in the left DLPFC, and the left NAcc, as well as the intrinsic FC of the DLPFC-NAcc between early improvers and non-improvers. We evaluated the association between the baseline brain imaging features (ALFF, ReHo, and FC) and improvements in depression and anxiety symptoms. We found that the pretreatment ALFF and ReHo in the stimulated DLPFC and left NAcc did not significantly differ between the subgroups. The early improvers displayed increased negative FC strength between the stimulated DLPFC and left NAcc with respect to non-improvers. The stimulated DLPFC-NAcc FC strength negatively correlated with improved depressive and anxious symptoms. This study is the first to demonstrate that the resting-state FC of the stimulated DLPFC-NAcc, rather than regional brain activity or local synchronization in the stimulated target, might predict the anti-depression and anti-anxiety effects of rTMS for depression.
