Targeted re-sequencing of the bovine collagenous lectins and peptidoglycan recognition proteins

ABSTRACT: This study targeted the bovine collagenous lectins, ficolins, MASP genes, and peptidoglycan recognition proteins of cattle. Hepatic DNA was sourced from 120 cattle presenting for postmortem examination at the University of Guelph. The animals were divided into pools of 5 animals each based on the similarity of postmortem diagnosis and all pools were sequenced on an Illumina MiSeq. Variants were called using GATK and allele frequency analysis performed using Popoolation2.

The significance of BIV infection in cattle has not been clearly established and it is still unknown whether BIV induces a specific syndrome or whether it renders animals more susceptible to other infections. To gain insight into host response to BIV infection the pattern of gene expression in bovine macrophage cells (BoMac) was analyzed using BLO Plus microarrays from CAFG, Michigan State University ( GEO GPL9176).

Transcriptional profiling of disease progression in bovine tuberculosis and the identification of potential diagnostic biomarkers

Project description:The definition of biomarkers correlating with disease progression could have impact on the rational design of novel diagnostic approaches for bovine tuberculosis in cattle. We have used the advantages of the murine model to identify promising candidates in the host transcriptome post-infection. RNA from BALB/c splenocytes and lung cells after aerogenic Mycobacterium bovis infection were with high-density microarrays to defining biomarkers of disease progression. In antigen-stimulated splenocytes we found statically significant modulation of 1109 genes early after infection and 1134 at later time-point post-infection. 618 of these genes were modulated at both time points. In the lung 282 genes were significantly modulated post-infection. Amongst the most strongly up-regulated genes were granzyme A in spleen, and cxcl9, granzyme B, interleukin-22, interluekin-17A receptor, and ccr6 in lungs. The expression of 20 of the most up-regulated genes identified in the murine studies were evaluated using PBMC from uninfected and naturally infected cattle. We could show that the expression of cxcl9, granzyme A and interleukin-22 following in vitro stimulation with PPD was significantly increased in infected cows compared to naïve animals. Thus, we have demonstrated that murine transcriptome analysis can be used to predict responses in cattle. In addition, we have prioritised CXCL9, GnzA and IL-22 as potential additional readout systems for the ante-mortem diagnosis of bovine tuberculosis.

Transcription profiling of Bos taurus surgical model of the jejunum infected with Bovine rotavirus (BRV) and bovine coronavirus (BCV)

Project description:Bovine rotavirus (BRV) and bovine coronavirus (BCV) infect intestinal villous epithelium in young cattle. A surgical model was adapted for neonatal calves in which a region of the jejunum was isolated from the digestive tract but lymph drainage, enervation and blood flow were maintained. Replicate sections of intestine (loops) were infected with either BRV or BCV and adjacent segments were injected with phosphate-buffered saline. Tissues were collected 18 hours post-infection. Four animals were used for BRV infection studies, and three animals were used for BCV infection studies. Microarray analyses provided a global evaluation of host gene expression patterns following BRV and BCV infection and changes in gene expression were validated by qRT-PCR analyses.

Targeted Transcriptome Analysis of Beef Cattle Persistently Infected with Bovine Viral Diarrhea Virus

Project description:Bovine Viral Diarrhea Virus (BVDV) is an endemic virus of North American cattle populations with significant economic and animal health impacts. While BVDV infection has a myriad of clinical manifestations, a unique and problematic outcome is the establishment of a persistently infected (PI) animal following in-utero viral infection. While it is well established that PI animals serve as a constant reservoir of BVDV, the mechanism for the maintained infection remains unknown despite multiple theories. The purpose of this study was to use transcriptome analysis to further define long term immune status of adult PI cattle and offer insight into the potential mechanistic establishment of persistent BVDV infection, in utero. Peripheral blood mononuclear cells were collected from PI beef cattle (N=6) and uninfected controls (N=6) for targeted RNAseq analysis conducted using 54 genes of interest and followed by pathway enrichment analysis. Analysis revealed 29 differentially expressed genes (FDR < 0.05, fold change > 2) representing 14 significant KEGG pathways between PI and control animals (FDR < 0.05). Transcriptome changes indicate chronic upregulation of interferon gamma (IFNG) with unexpected expression of related genes, suggesting a maintained stimulation of the PI immune system resulting in virus-mediated dysregulation of immune function.

Global gene expression of B. abortus-infected bovine macrophages

Project description:In a time course study, we characterized global gene expression profile of B. abortus-infected macrophages from cattle naturally resistant (R) and susceptible (S) to brucellosis. B. abortus infection causes early down-regulation of transcript levels in Mø from R cattle at 4 h p.i. (22 up- and 126 down-regulated genes) which is reversed by 12 h post-infection (31 up- and 25 down-regulated genes), compared to uninfected control. On the other hand, B. abortus-infected S bovine macrophages exhibited a down-regulated expression profile at 4 (45 up- and 65 down-regulated genes) and 12 h p.i. (47 up- and 193 down-regulated genes). The analysis of the results indicates that B. abortus – infected Mø from cattle naturally R and S to brucellosis display different transcriptional profiles. Specific genes and biological processes identified in this study will further help elucidate how different macrophages from resistant and susceptible animals interact with Brucella during the early infectious process. Using the Affymetrix Bovine Genome Array, global mRNA expression analysis of immature (GV) and in vitro matured (IVM) bovine oocytes was carried out to characterize the transcriptome of the bovine oocyte and to identify the key pathways associated with oocyte meiotic maturation and developmental potential.
