The profile of exosomal and cellular miRNA in ovarian cancer cell lines

ABSTRACT: To check the profile of exosomal and cellular miRNA in ovarian cancer cell lines, total RNA were extracted from exosomes and cells. Thirteen ovarian cancer cell lines (A2780, ES-2, CAOV3, SKOV3, OV-90, OAW42, MCAS, COV362, RMG-1, RMUG-S, KURAMOCHI, NIH-OVCAR3 and A2780cis) were investigated, and HOSE1, HOSE2 and HOSE3 (human ovarian surface epithelim cell lines) were used as control.

Project description:Analysis of a panel of 41 human ovarian cancer cell lines for the study of ovarian cancer biology, drug response and drug sensitivity. Ovarian cancer cell lines were either obtained from the American Type Culture Collection (ATCC, Manassas, VA) (CAOV3, OV90, OVCAR3, SKOV3, TOV112D), the European Collection of Cell Cultures, Salisbury, England (A2780CP, A2780S); Kyoto University, Kyoto, Japan (CHI, CHIcisR, M41, M41CSR, Tyknu, and TyknuCisR), or were kind gifts from Dr. Patricia Kruk, Department of Pathology, College of Medicine, University of South Florida, Tampa, FL; and Susan Murphy, PhD, Dept of OBGYN/Division of GYN Oncology, Duke University, Durham, NC (A2008, C13, CAOV2, FUOV1, HeyA8, IGR-OV1, IMCC3, IMCC5, MCAS, OV2008, OVCA420, OVCA429, OVCA432, OVCA433, OVCAR4, OVCAR5, OVCAR8, Dov13, BG1, Ovary1847, OVCAR10, OVCAR2, SK-OV-4). Cell lines were maintained in RPMI-1640 (Invitrogen; Carlsbad, California) supplemented with 10% fetal bovine serum (Fisher Scientific; Pittsburg, PN), 1% sodium pyruvate, 1% penicillin/streptomycin (Cellgro; Manassas, VA), and 1% nonessential amino acids (HyClone; Hudson, New Hampshire). Mycoplasma testing was performed every six months following manufacturer’s protocol (Lonza, Rockland ME).

Project description:In order to know whether fibroblast-derived exosomes affect cancer cell behavior, we sprinkledtreated those onto CaOV3 and OVCAR3, and analyzed those effects using gene expression microarrays. The key enzyme of Lewis y synthesis is human fucosyltransferase gene 1(hfut1). Through transfecting hfut1 into Lewis y low expressing cell line RMG-I, we got a pair of cell lines. We hypothesized that Lewis y antigen plays a major role in ovarian canceration . Using gene arrays, we comprehensively profiled mRNA differential expression between fut1-pretransfected and posttransfected clear cell of human epithelial OvCa. We expected that over-expression of lewis y antigen would lead to higher expression of genes associated with growth, metastasis,proliferation and so on. we established ovarian tumor cell line RMG-I-H by stably transfecting human fucosyltransferase gene 1 into cell line RMG-I. RMG-I-H overexpresses Lewis y antigen, which is known as a cancer-related tetrasaccharide antigen. We profiled for gene expression both cells pretransfected and posttransfected.

Wogonin inhibitsovarian cancer by activating the AMPK-TET2-5hmC axis

Project description:Ovarian cancer is one of the most common female cancers. In the quest of effective anti-cancer agents, this study explores the effects of wogonin, a naturally occurring flavonoid, on the viability and migration of A2780 and Kuramochi ovarian cancer cell lines. Our findings demonstrate that wogonin not only impedes cancer cell growth and mobility in both in vitro and in vivo but also significantly enhances the cytotoxic efficacy of cisplatin. Mechanistic investigations reveal that wogonin suppresses genes associated with proliferation and EMT and upregulates metabolic pathways, particularly the AMPK signaling, which is crucial for increasing 5hmC levels, highlighting wogonin's role in promoting DNA demethylation through the stabilization of TET2. Our findings not only highlights the therapeutic potential of wogonin but also its preventative capability against ovarian cancer in individuals with metabolic disorders, such as diabetes, who are at increased risk for ovarian cancer. This study highlights wogonin's dual functionality in ovarian cancer treatment and prevention, emphasizing its promising prospects for future oncological applications.

Expression changes following siRNA knockdown of ADRM1 in ovarian cell line OAW42

Project description:Knock-down of ovarian cancer amplification target ADRM1 leads to down regulation of GIPC1 and up-regulation of RECK. Among 20q13-amplified genes in ovarian cancer, ADRM1 overexpression was the most highly correlated with amplification, and was significantly upregulated with respect to stage, recurrence and metastasis. In addition, overexpression of ADRM1 correlated significantly with shorter time to recurrence and overall survival. Herein, array-CGH and microarray expression of ovarian cancer cell lines, provides evidence consistent with the primary tumor data that ADRM1 is a 20q13 amplification target. Knock-down of ADRM1 in amplified ovarian cell line OAW42 results in down-regulation of growth factor GIPC1 and up-regulation of tumor-suppressor RECK RNA and protein. In our dataset of 141 ovarian primary tumors, ADRM1 overexpression significantly correlates with GIPC1 overexpression. In addition, there is a significant anticorrelation between ADRM1 overexpression and RECK expression. Further research is necessary to determine whether targeting knock-down of ADRM1 in 20q13-amplified ovarian cancers results in growth inhibition and tumor suppression via downstream targets GIPC1 and RECK.
