Project description:We performed genome-wide profiling experiments on tissues from Drosophila melanogaster adult females with a mutation in the p24 gene logjam (loj) to test the hypothesis that loss of loj function causes a transcriptional response characteristic of ER stress activation. Experiment Overall Design: We compared expression profiles from loj mutant females to those of controls. We compared mutant and control abdomen tissue as well as mutant and control tissue preparations from the remainder of the fly (the head/thorax). Three mutant and three control RNA samples for each set of tissue were used for array hybridization (12 total array samples).

Project description:Wild-type (C57Bl6) mice underwent monocular enucleation at P20 and were sacrificed 4 days later at P24. Bilateral (non-deprived and deprived) visual cortex was dissected from 1-mm thick coronal sections, and total RNA was extracted from the tissue lysate using Trizol (Gibco-BRL). Experiment Overall Design: Gene expression in pooled samples from 4 non-deprived (ipsilateral to monocular enucleation) and 4 deprived (contralateral to monocular enucleation) visual hemispheres was compared.

Females translate male mRNA transferred during mating

Project description:Although the presence of RNA in seminal fluid appears to be a conserved feature of male ejaculates, it is unknown whether this RNA is functional within females. Here, we develop an experimental proteomic method called VASPA (Variant Assisted SILAC Proteomic Analysis) to test the hypothesis that Drosophila male seminal fluid RNA is translated by females. We find evidence for 166 male-derived, female-translated proteins (mdFTPs) in female lower reproductive tracts at six hours postmating, many with predicted functions relevant to reproduction.
