Results: Muscle transcriptomic profiles demonstrate a prominent signature of mitochondrial dysfunction in sarcopenic patients. Conclusions: Our study supports the fundamental role of mitochondrial dysfunction in driving pathological muscle and mobility decline in the elderly.

Single-nuclei profiling of aged skeletal muscle

Project description:Aging is accompanied by a loss of muscle mass and function, termed sarcopenia, causing numerous morbidities and economic burdens in human populations. Mechanisms implicated in age-related sarcopenia include inflammation, muscle stem cell depletion, mitochondrial dysfunction and loss of motor neurons, but whether there are key drivers of sarcopenia is not yet known. To gain deeper insights into age-related sarcopenia, we performed transcriptome profiling on lower limb muscle biopsies from 72 young, old and sarcopenic subjects using bulk RNA-seq (N = 72) and single-nuclei RNA-seq (N = 17). Using this combined approach, we discovered novel changes in gene expression that occur with age and sarcopenia in multiple cell types comprising mature skeletal muscle. Notably, we found increased expression of the genes MYH8 and PDK4, and decreased expression of the gene IGFN1, in old muscle. We also identified a small population of nuclei that express CDKN1A, present only in aged samples, consistent with P21CIP1 senescence in this subpopulation. Our findings identify unique cellular sub-populations populations in aged and sarcopenic skeletal muscle, which will facilitate the development of new therapeutic strategies to combat age-related sarcopenia.

Mouse Skeletal Muscle Sarcopenia

Project description:Our study aimed to characterize sarcopenia in C57BL/6J mice using a clinically relevant definition and age range and investigate the underlying mechanisms leading to sarcopenia. Aged male C57BL/6J mice (23-32 months) were classified as non-sarcopenic (no deficit), probable sarcopenic (1 deficit), and sarcopenic (2-3 deficits) based on assessments of grip strength, muscle mass, and treadmill running time and using 2 standard deviations below the mean of the young (4-9 months) as cut-off points. A 9-22% prevalence of sarcopenia was identified in 23-26-month-old mice. Age-related declines in muscle function were more severe than in muscle mass and outcomes of all three assessments were positively correlated in aged mice. As sarcopenia progressed, there were decreases in specific force as well as fiber size and number of IIB skeletal muscle fibers. Mitochondrial biogenesis, oxidative capacity, and AMPK-autophagy signaling decreased significantly while no increase in atrogenes was detected. Our study is the first to characterize sarcopenia in C57BL/6J mice using a clinically relevant definition and highlights the different trajectories of age-related declines in muscle mass and function. These critical insights into the molecular changes associated with sarcopenia progression will facilitate future development of therapeutic interventions.

Epigenome analysis of muscle tissue samples from male participants of the Hertfordshire Sarcopenia Study (HSS) and Hertfordshire Sarcopenia Study extension (HSSe)

Project description:Genome wide DNA methylation profiling of muscle tissue genomic DNA. The Illumina Infinium MethylationEPIC array was used to obtain DNA methylation profiles across approximately 850,000 CpGs. Samples were male, taken from both the Hertfordshire Sarcopenia Study (HSS, n=34) and the Hertfordshire Sarcoepnia Study exension (HSSe, n=44), to determine DNA methylation associations with measures of muscle mass and function. We further aimed to test whether differences in the muscle methylome are associated with sarcopenia in older individuals. We found significant associations between DNA methylation in the muscle tissue and sarcopenia, as well as individual measures of muscle mass and function, enriched around EZH2 binding sites.
