Project description:During the last decades, several mouse models provided insights into the regulation of folliculogenesis. In contrast to the commonly used transgenic or knockout mouse models, the Dummerstorf high-fertility mouse line 1 is a worldwide unique selection experiment for increased female reproductive performance and high fertility. These mice are characterized by various alterations on endocrine and molecular levels, which have the potential to improve folliculogenesis and therefore lead to increased ovulation rates. To get insights into the cellular mechanisms leading to the high fertility phenotype we used granulosa cells isolated from antral follicles for mRNA sequencing and measured hormones and growth factors associated with follicular development. Our results indicate that the crosstalk between different intracellular signaling pathways in the granulosa cells are improved, follicular atresia in FL1 might be decreased due to enhanced granulosa cell survival and by improving the efficiency of intracellular signaling, glucose metabolism and signal transduction, FL1 mice have several advantages in reproductive performance and therefore increased the ovulation rate without health issues.

Project description:As generally known, reproduction is referred to as a couple’s challenge. Many genes generally coding for reproduction performance are shared by both sexes. For investigating the impact of ‘high fertility’ breeding on murine males testis transcriptomic profile was examined of two worldwidely unique outbred long-term mouse lines which have been selected for large and heavy litters - Fertility Lines 1 and 2 (FL1, FL2); also known as DUK and DUC. After >170 generations of generally female focused breeding both high fertile mouse lines dramatically increased the number of offspring as well as the total litter weight up to 180%, which is additionally accompanied by raised ovulation rate as could be shown by previous data. Concomitantly with the improved fertility phenotype, for the FL males several line-specific phenotypic characteristics could already be revealed like physiological parameters (e.g. higher bodyweight, accelerated puberty and life expectancy), behavioural parameters (e.g. explorative behaviour in an open field and decreased endurance fitness on a treadmill) or testicular cell type composition (e.g. altered haploid and diploid cell content shown by flow cytometric analysis). By this combination of phenotypic characterization and profound transcriptomic profiling we hope to ensure for future perspective that these genetically heterogeneous mouse models will provide new insights into molecular and cellular mechanisms of enhancing male fertility.

Project description:The aim of this study was to assess the impact of oocyte competence on subsequent fertility. Based on knowledge already accessible in mammals and on bioinformatics tools including the chicken genome sequence, we focused on the expression of genes involved in the processes of fertilization and of early embryo development. A differential kinetic study is performed on INRA lines selected on the basis of their fertility potential in purpose of hopefully access gene markers of fertility performance. We use 4 different hen lines:; - one line of laying hens with 3 different samples: the just ovulated oocyte, the oocyte collected 24 hours before ovulation (F1 stage), and granulosa cells collected at the F1 stage. We could compare different tissue and developmental stages. - one line of hen with rapid growth speed; - two lines of laying hens; For the 3 last lines we used animals with different fertility levels. We collected the oocyte of the largest follicle before ovulation (F1). The aim of the study is to identify genes involved in fertility or early embryo mortality. Project description:BackgroundIn a recent study it was demonstrated that a largely increased ovulation number is responsible for high prolificacy in two mouse lines selected for fertility performance. The objective of the present study was to identify genes that are involved in increasing the ovulation number in one of these lines, FL1. For differential expression profiling, ovaries of FL1 and of a non-selected control line, DUKsi, both lines derived from the same genetic pool, were analyzed with microarray analysis and quantitative polymerase chain reaction (qPCR). Ovaries from 30 animals of each line were collected at the metestrous stage, combined to 6 pools each, and processed for microarray analysis.ResultsThe actual number of ova shed in FL1 exceeded that of the DUKsi control line more than twofold (26.6 vs. 12.9). 148 differentially expressed ovarian transcripts could be identified, 74 of them up- and 74 down-regulated. Of these, 47 significantly mapped to specific Gene Ontology (GO) terms representing different biological processes as steroid metabolism, folliculogenesis, immune response, intracellular signal transduction (particularly of the G protein signaling cascade), regulation of transcription and translation, cell cycle and others. qPCR was used to re-evaluate selected transcripts and to estimate inter-individual variation of expression levels. These data significantly correlated with microarray data in 12 out of 15 selected transcripts but revealed partly large variations of expression levels between individuals.Conclusion(1) The abundance of numerous ovarian transcripts was significantly different in FL1 compared to the non-selected control line DUKsi thus suggesting that at least some of the respective genes and corresponding biological processes are involved in improving reproductive traits, particularly by increasing the number of ovulation. (2) Selective qPCR re-evaluation largely confirmed the microarray data and in addition demonstrated that sample pooling can be beneficial to find out group-specific expression profiles despite of large inter-individual variation. (3) The present data will substantially help ongoing genetic association studies to identify candidate genes and causative mutations responsible for increased fertility performance in mice.

Project description:Single-cell RNA-seq (10x) of the goat ovarian granulosa cells samples from high fertility group (>three babys per birth) and low fertility group (< two babys per birth). The goal of this project is to explore the role of ovarian granulosa cells on the fertility of goat at single cell level. We would like to investigate the role of ovarian granulosa cells on the ovulation rate which indicates fertility. Therefore the ovarian granulosa cells from different population (hihg or low fertility groups animals) were analyzed.
