Flow-Mediated Modulation of the Endothelial Cell Lipidome

Project description:The luminal surface of the endothelium is exposed to dynamic blood flow patterns that are known to affect endothelial cell phenotype. In this study, human aortic endothelial cells (HAECs) were exposed to unidirectional laminar flow (20 dynes/cm2) or disturbed flow (±4 dynes/cm2, 2 Hz) for 48 hrs and then subjected to RNA sequencing. The sequencing data was used to analyze changes in endothelial cell gene expression and pathways associated with lipid metabolism.

Project description:This study investigates the role of endothelial cell (EC) gene expression in the focal origin of atherosclerosis, particularly in response to local hemodynamics. Differential gene expression was profiled in EC isolated from athero-susceptible and athero-protected regions of the normal pig aorta. Specifically, a region of disturbed flow (DF, the inner aortic arch) was compared to a region of undisturbed flow (UF, descending thoracic aorta). Linearly amplified RNA was used to screen nylon filter arrays of 13,824 human cDNAs.

The role of MAGI1 (membrane-associated guanylate kinase with inverted domain structure-1) in regulating disturbed flow-induced changes of gene expression

Project description:To understand the function of MAGI1 in ECs, we isolated total RNAs from human umblical vein endothelial cells (HUVECs) transfected with control or Magi1 siRNA and transcriptionally profiled them. Ingenuity pathway Analysis (IPA) demonstrated activation of antiviral responses, including interferon signaling, without affecting NF-κB expression and inhibition of ER stress gene expression induced by the reduction of MAGI1 expression. Because activation of interferon signaling tends to increase NF-κB activation, the finding that depletion of MAGI1 promotes antiviral responses without increasing NF-κB activation is unique, suggesting that MAGI1 is an incomparable molecular switch for pro-viral and pro-inflammatory responses.

Transcription profiling of human aortic endothelial cells with miR-10a knock down

Project description:This study investigates the miR-10a-mediated transcriptome in human aortic endothelial cells (HAECs). The EC transcriptome was profiled in miR-10a knockdown HAECs. Specifically, small amounts of EC RNA were isolated from control cells and miR-10 knockdown cells expressing miRNA inhibitor control (Dharmacon) and miR-10a inhibitors (Dharmacon), respectively . Evidence for the importance of OxPAPC interaction with cysteines in regulating endothelial cell function

Project description:Oxidation products of 1-palmitoyl-2-arachidonoyl-sn-glycerol-2-phosphatidylcholine (PAPC), referred to as OxPAPC, accumulate in atherosclerotic lesions and regulate over 1000 genes in human aortic endothelial cells (HAEC). We previously demonstrated that OxPAPC covalently binds to a number of proteins in HAEC. The goal of these studies was to gain insight into the binding mechanism and determine if binding regulates activity. In whole cells N-acetylcysteine inhibited gene regulation by OxPAPC, and blocking cell cysteines with N-ethylmaleimide strongly inhibited OxPAPC binding. Using MS, we demonstrate that most of the binding of OxPAPC to cysteine is mediated by PEIPC. We also show that OxPAPC binds to a model protein, H-Ras, at cysteines previously shown to regulate activity in response to 15dPGJ2. This binding was observed with recombinant protein and in cells overexpressing H-Ras. 15dPGJ2 and OxPAPC increased H-Ras activity at comparable concentrations. Using microarray analysis, we demonstrate a considerable overlap of gene regulation by OxPAPC and 15dPGJ2 in HAEC, suggesting that some effects attributed to 15dPGJ2 may also be regulated by OxPAPC since OxPAPC lipids and 15dPGJ2 both accumulate in inflammatory sites. Overall, we provide evidence for the importance of OxPAPC-cysteine interactions in regulating HAEC function. Control, OxPAPC, and 15dPGJ2-treated HAECs Samples. 'Control' is media (1%FBS, 99%M199)-only treated HAECs. 15dPGJ2 and OxPAPC are treated with those respective compounds in media.
