ABSTRACT: Gene expression was determined for normal human bronchial epithelial (NHBE) cells incubated with DMSO or TG immediately prior to infection with USSR H1N1 virus for 12 hours. All of the NHBE cell stocks were obtained from QIAGEN.

Expression data from well-differentiated human bronchial epithelial cells infected with H1N1 Influenza isolates

Project description:We used microarrays to compare the gene expression profiles of different H1N1 isolates (seasonal and pandemic) in lung epithelial cells in vitro. Well-differentiated primary human lung, bronchial epithelial cells (wd-NHBE) were infected with two pandemic and one seasaonl H1N1 influenza viruses at a multiplicity of infection of 3.0. The cells were then collected, in triplicate, at 36 hours after infection. This timepoint was selected as we saw significant differences in soluble cytokine and chemokine protein levels at this time point in the supernatant.

RNA sequencing of human bronchial epithelial cell transcriptomes after repopulation and differentiation on bronchial scaffolds from COPD patients and healthy donors

Project description:Chronic obstructive pulmonary disease (COPD) is a serious global health problem characterized by chronic airway inflammation, progressive airflow limitation and destruction of lung parenchyma. Remodeling of the bronchial airways in COPD includes changes in both the bronchial epithelium and the subepithelial extracellular matrix (ECM). To explore the impact of an aberrant ECM on epithelial cell phenotype in COPD we developed a new ex vivo model, in which normal human bronchial epithelial (NHBE) cells repopulate and differentiate on decellularized human bronchial scaffolds derived from COPD patients and healthy individuals. By using transcriptomics, we show that bronchial ECM from COPD patients induces differential gene expression in primary NHBE cells when compared to normal bronchial ECM. The gene expression profile indicated altered activity of upstream mediators associated with COPD pathophysiology, including hepatocyte growth factor, transforming growth factor beta 1 and platelet-derived growth factor B, which suggests that COPD-related changes in the bronchial ECM contribute to the defective regenerative ability in the airways of COPD patients.

Ca2+ entry modulates oscillation frequency by triggering Ca2+ release.

Project description:As in many cells, the frequency of agonist-induced cytosolic Ca2+ concentration ([Ca2+]1) oscillations in exocrine avian nasal gland cells is dependent on the rate of Ca2+ entry. Experiments reveal that the initiation of each oscillatory spike is independent of the relative fullness of the stores and, furthermore, the oscillating pool is normally fully refilled by the end of each [Ca2+]1 spike. Therefore, contrary to current models, the interspike interval (which essentially sets the frequency) does not reflect the time taken to recharge the oscillating stores. Instead, the data show that it is the previously demonstrated role that Ca2+ entry plays in triggering the repetitive release of Ca2+ from the oscillating stores, rather than the recharging of those stores, that provides the basis for the observed effects of Ca2+ entry rate on oscillation frequency.

Analysis of the apical secretome of Calu-3 and NHBE cells in 3D models of the human bronchial epithelium

Project description:The mucus secreted by airway epithelial cells plays a key role in the protection against and clearance of particles and pathogens. In this project, a 3D model of the bronchial epithelium was established using Calu-3 cells grown on porous inserts at the air-liquid interface. The secreted proteins were characterized in long-term cultures and compared to the apical secretome of primary normal human bronchial epithelial (NHBE) cells. The apical secretome was collected and characterized in the Calu-3 model at day 4, day 11, and day 18 after air-liquid interface and in the NHBE model at day 4, day 12, and day 18 after air-liquid interface. Ca2+ entry following P2X receptor activation induces IP3 receptor-mediated Ca2+ release in myocytes from small renal arteries.

Project description:P2X receptors mediate sympathetic control and autoregulation of the renal circulation triggering contraction of renal vascular smooth muscle cells (RVSMCs) via an elevation of intracellular Ca(2+) concentration ([Ca(2+) ](i) ). Although it is well-appreciated that the myocyte Ca(2+) signalling system is composed of microdomains, little is known about the structure of the [Ca(2+) ](i) responses induced by P2X receptor stimulation in vascular myocytes.Using confocal microscopy, perforated-patch electrical recordings, immuno-/organelle-specific staining, flash photolysis and RT-PCR analysis we explored, at the subcellular level, the Ca(2+) signalling system engaged in RVSMCs on stimulation of P2X receptors with the selective agonist ??-methylene ATP (??-meATP).RT-PCR analysis of single RVSMCs showed the presence of genes encoding inositol 1,4,5-trisphosphate receptor type 1(IP(3) R1) and ryanodine receptor type 2 (RyR2). The amplitude of the [Ca(2+) ](i) transients depended on ??-meATP concentration. Depolarization induced by 10?µmol·L(-1) ??-meATP triggered an abrupt Ca(2+) release from sub-plasmalemmal ('junctional') sarcoplasmic reticulum enriched with IP(3) Rs but poor in RyRs. Depletion of calcium stores, block of voltage-gated Ca(2+) channels (VGCCs) or IP(3) Rs suppressed the sub-plasmalemmal [Ca(2+) ](i) upstroke significantly more than block of RyRs. The effect of calcium store depletion or IP(3) R inhibition on the sub-plasmalemmal [Ca(2+) ](i) upstroke was attenuated following block of VGCCs.Depolarization of RVSMCs following P2X receptor activation induces IP(3) R-mediated Ca(2+) release from sub-plasmalemmal ('junctional') sarcoplasmic reticulum, which is activated mainly by Ca(2+) influx through VGCCs. This mechanism provides convergence of signalling pathways engaged in electromechanical and pharmacomechanical coupling in renal vascular myocytes.
