Tumor-infiltrating immune cells of young and aged mice

ABSTRACT: Macrophages, dendritic cells, conventional CD4+ T cells, CD8+ T cells, and regulatory T cells isolated from mouse colon cancer model MC38 tumors implanted subcutaneously to young (3 month) and aged (12 month) mice were sequenced using ImmGen's standard ultra-low input RNA-seq pipeline, in order to study age-dependent differences in intraltumoral immune cell functions and their impact on tumor control

Project description:Obesity is a major cancer risk factor, but the underlying molecular mechanisms are not always known. In this study, we look at proteome remodeling in cancer cells with obesity, comparing tumor cells sorted from mice fed high-fat versus control diet. We conducted 10-plex TMT-proteomics on GFP+ MC38 colorectal adenocarcinoma tumor cells, sorted from subcutaneously implanted tumors 12 days after implantation. This study reveals molecular pathways that change in cancer cells with obesity that promote tumor growth.

Project description:CT26 tumors were implanted subcutaneously into syngeneic BALB/C mice and allowed to grow for 15-25 days. Tumors were collected, mechanically dissociated, and immune cells enriched using Percoll gradient. Cells were then stained with viability dye, CD45, CD3, and NKp46 to FACS sort for T cells (Live/CD45+/CD3+/NKp46-) and NK cells (Live/CD45+/CD3-/NKp46+). Isolated tumor infiltrating NK and T cells were then processed for RNA sequencing analysis.

Considering PB-020’s good pharmacokinetic properties, combined effect of PB-020 and immunotherapy drugs is worth studying. As PB-020 can effectively suppress mouse colorectal cancer cell lines in cell culture, we tested the effect of combining PB-020 with the anti-PD-1 monoclonal antibody RMP1-14 on the growth of subcutaneously implanted MC38 cells to C57BL/6 mice as a syngeneic mouse model of colorectal cancer. To clarify the molecular mechanism underling this anti-cancer therapy, we executed RNA sequencing of tumor sample obtained from the experiment above.

Project description:Skeletal stem and progenitor cells (SSPCs) perform bone maintenance and repair. With age, they produce fewer osteoblasts and more adipocytes leading to a loss of skeletal integrity. The molecular mechanisms that underlie this detrimental transformation are largely unknown. To identify transcriptional changes that occur with aging we performed bulk RNA sequencing on young (3 month) and middle-aged (12 month) SSPCs.

Project description:We performed MIcroarray experiments on a total of 12 immune organs, including spleen (SP), bone marrow (BM), lymph node (LN) and Peripheral blood mononuclear cell (PBMC) and three tumor tissues including MC38, EL4 and LLC. 12 mice from four immune organs, including the spleen (SP), bone marrow (BM), lymph nodes (LN), and Peripheral blood mononuclear cell (PBMC), were isolated using routine experimental methods.9 mice were randomly divided into 3 groups and implanted subcutaneously(s.c.) with EL4 (2.0*10^5 /mice), MC-38 (1.0*10^6 /mice), and LLC-JSP-t2 (2.0*10^5 /mice) cells, respectively. All mice were sacrificed and tumor tissues were collected when the tumor volume reached 100 mm 3 . The tissues were ground up with liquid nitrogen and the lysates were stocked using Trizol for subsequent array analysis. RNA isolation and hybridization were performed by Capitalbio Technology Corporation according to the standard Affymetrix protocol .
