Project description:Newborn mice were concomitantly treated with antagomiR-scrambled (SCR) or antagomiR-34a (A34a) and exposed either to normoxia (21% O2) or hyperoxia (85% O2). Lungs were harvested at two different time-points, P5.5 and P14.5, in order to compare the differential mRNA expression between the 4 groups. Unfortunately, few changes in mRNA expression were detected.

Silencing miR-31 in naive CD4+ T cells by antagomiRNA

Project description:To describe the global profile of genes regulated by miR-31, we compared the transcriptomics between miR-31 silenced and antagomir control treated naive CD4+ T cells. Interestingly, the cells could be separated into two subsets according FCS and SSC by flow cytometer after the silence of miR-31, one (~65%) was small sized cells with low granularity, the other (~30%) was large cells with high granularity, whereas the antagoNC treated naïve CD4+ T cells only contain a marginal fraction of large cells. We pooled “antagoNC”, “antagomiR31S” and “antagomiR31L” naïve CD4+ T cells from 5 healthy donors, respectively, and analyzed the differential genes among those three subsets.

Project description:Transcriptional profiling of infrarenal aortic tissue from Male 10-week-old C57BL/6J mice after AAA-induction with porcine pancreatic elastase, compared with sham-operated saline-injected mice. One day after AAA-induction, the mice were injected intraperitoneally with either lentiviral packaged miR-24 antagomir (anti-miR-24) or miR-24 mimic (pre-miR-24), or a scrambled microRNA control (scr-miR). Aortic samples were obtained 7 days after operation. The goal was to examine gene expression in developing AAA in this model, and to compare the effects of scr-miR, anti-miR-24 and pre-miR-24. Four condition experiment, one infrarenal aorta per array. Sham vs. scr-miR-PPE vs. anti-miR-24-PPE vs. pre-miR-24-PPE, all harvested at Day 7 post-operatively. After QC, the final analysis group (uploaded here) consisted of 18 arrays: Sham-Saline-treated (4 arrays), scr-miR-PPE-treated (5 arrays); pre-miR-24-PPE-treated (3 arrays); and anti-miR-24-PPE-treated (6 arrays).

Study of the role of miR-205 in prostate epithelial cells

Project description:Gene expression has been assessed on RWPE-1 cells, derived from normal prostate, upon silencing of miR-205 by an antisense locked nucleic acid. RWPE-1 cells were transfected in triplicate either with 100nM of an antisense locked nucleic acid targeting miR-205 (LNA-205) or a scrambled oligomer (LNA-Scr). miRNAs affected by antagomiR-17 treatment

Project description:To understand the the effect of antagomir-17 treatment on human endothelial cells derived from human umbilical cord blood (UCB) CD34+ hematopoietic stem cells, we have employed mRNA sequencing. The antagomiR-17 used in this study was purchased from Dharmacon and cell transfection was performed using Lipofectamine RNAiMAx from Life Technologies. Scramble antagomiR from Ambion was used as control. Cells were transfected with antagomiR-17 or scrambled antagomiR for 48 hours. After 48 h, the cells were collected, RNA was isolated and RNA samples were shipped to Exiqon Services, Denmark for mRNA sequencing. All sequencing experiments (RNA integrity measurements, library preparation and next generation sequencing) were conducted at Exiqon Services, Denmark.
