Shift work disrupts circadian regulation of the transcriptome in hospital nurses

ABSTRACT: In the present study, we sought to determine the degree of circadian misalignments of hormonal and transcriptional rhythms with the timing of sleep-wake behavior on days off in day-shift and night-shift hospital nurses. We conducted a genome-wide microarray analysis on RNA isolated from PBMCs to examine individual variability of transcriptional rhythms.

The effect of simulated night shift work on the circadian regulation of the human transcriptome

Project description:Eight healthy human subjects were enrolled in a 6-day simulated shift work protocol. Blood samples were collected during the two 24-hour measurement periods. Blood samples were collected every 4 hours during both measurement periods. Subjects entered the lab on Day 1. At the start of Day 2, the first 24-hour measurement period was started. Subjects slept according to their habitual sleep/wake schedule, followed by a 16-hour constant posture procedure. On days 3-6, the sleep period was delayed by 10 hours. Following the third night on this schedule, subjects underwent another 24-hour measurement period. During both measurement periods, 7 blood samples were collected and PBMCs were isolated. mRNA was extracted, labelled, and hybridized to microarrays.

Sleep-wake cycles drive daily dynamics of synaptic phosphorylation

Project description:The circadian clock drives daily changes of physiology, including sleep-wake cycles, by regulating transcription, protein abundance and function. Circadian phosphorylation controls cellular processes in peripheral organs, but little is known about its role in brain function and synaptic activity. We applied advanced quantitative phosphoproteomics to mouse forebrain synaptoneurosomes isolated across 24h, accurately quantifying almost 8,000 phosphopeptides. Remarkably, half of the synaptic phosphoproteins, including numerous kinases, had large-amplitude rhythms peaking at rest-activity and activity-rest transitions. Bioinformatic analyses revealed global temporal control of synaptic function via phosphorylation, including synaptic transmission, cytoskeleton reorganization and excitatory/inhibitory balance. Remarkably, sleep deprivation abolished 98% of all phosphorylation cycles in synaptoneurosomes, indicating that sleep-wake cycles rather than circadian signals are main drivers of synaptic phosphorylation, responding to both sleep and wake pressures.

Multiple brains were pooled for each sample and used for the genome-wide DNA methylation analysis.

Mistimed sleep disrupts circadian regulation of the human blood transcriptome

Project description:22 healthy volunteers without sleep disorders were resident in an environmental scheduling facility and participated in a forced-desynchrony protocol, in which the sleep-wake cycle and the associated fasting-feeding cycle is scheduled to a 28-hour period, of which one third (i.e. 9h 20min) is scheduled for sleep. Under these conditions, during which light levels in the waking episode are kept low and sleep is scheduled in darkness, the phase of the melatonin rhythm occurred at approximately the same clock time during the first (D1) and fourth (D4) 28-h cycle and there were no major changes in either the amplitude or the waveform of this rhythm. During D1 and D4 7 blood samples were taken, RNA was extracted from leukocytes, labelled and hybridised to human whole-genome microarrays A total of 287 samples comprising 22 human subjects, for which 14 samples across multiple time-points/sleep condition were collected.

Proteomics of human peripheral blood mononuclear cells under normal circadian cycle and under a simulated night shift

Project description:Proteomic data from human peripheral blood mononuclear cells (PBMCs). Samples taken over a 24 hour constant light and diet following 3 days of normal day shift cycle or a night shift - circadian misaligned schedule where sleep and wake cycles were reversed. Time points (in clock time) 1, 7, 14, 21 hours; 6 volunteers for each condition. Samples were digested with trypsin, then analyzed by LC-MS/MS. Data was searched with MS-GF+ using PNNL's DMS processing pipeline.

Project description:Meal timing is essential in synchronization of circadian rhythms in different organ systems through clock-dependent and -independent mechanisms. The liver is a critical metabolic organ whose circadian clock and transcriptome can be readily reset by meal timing. However, it remains largely unexplored how circadian rhythms in the liver are organized in time-restricted feeding that intervenes meal timing. Here, we applied data-independent acquisition proteomics to characterize circadian features associated with day/sleep- (DRF) and night/wake (NRF)-time restricted feeding in nocturnal female mice.
