An integrated transcriptome analysis of immunological responses in pearl sac of pearl mussel Hyriopsis cumingii after mantle implantation

ABSTRACT: Our findings help us gain new insight into the immunological reactions induced by transplantation in pearl mussels and markedly bring us more clues about pearl sac formation, contributing to the novel development of management strategies for pearl culture.

Project description:To elucidate the modulatory participation of miRNAs in mollusk biomineralization, we have employed high-throughput sequencing to identify miRNAs of pearl oyster, Pinctada fucata. Our study focused on the miRNA expression profile of the mantle, an organ responsible for shell formation of the oyster. The pearl oysters were cultured in the tank with the maintaining conditions of temperature 19 ℃, PH 8.1 and salinity 33‰ in recirculating seawater.

Digital gene expression tag profile of five development stages of pearl sac from pearl oyster Pinctada martensii reveals potential candidates contributing to nacre formation

Project description:A sequencing depth of 5 million raw tags per sample was obtained and a large number of differentially expressed genes were identified. The results of the DGE tag profile were validated by examining gene expression in a subset of genes using qRT-PCR. Gene expression profiles at different pearl sac development stages implicated that a great change in the body of the host pearl oyster was induced by the surgical implantation and achieved the maximum at 15 days. An enrichment of genes relevant to purine metabolism was obtained by KEGG pathway analysis of the genes differentially expressed between 15 and 60 days. Analyzing gene expression trends, most of the immune-related genes, such as HSP70, TLR1 and lectin, were highly expressed before 30 days. While nacre formation-related genes, such as DPT and perlucin-like protein isoformA, and some immune-related genes, such as galectin and HSP90 like protein, were highly expressed at 60 days, indicating some immune-related gene may participate in nacre formation. Furthermore, we found 286 unigenes highly expressed at 60 days. These unigenes may be potential candidates contributing to nacre formation, such as leucine-rich repeat-containing protein, secreted protein with cysteine rich repeats and Cathepsin O.

The mantle exosome and microRNAs of Hyriopsis cumingii involved in nacre color formation

Project description:The nacre color of shells has an effect on the pearl color in Hyriopsis cumingii, and is an important indicator for its value. However, little exosome and micro (mi)RNA information are available on nacre color formation in mussels. In this study, exosomes of mantles were extracted from white and purple mussels. High-throughput Illumina sequencing was performed on the white and purple mussel mantle exosomes.Moreover, miR-223 negatively regulated hcApo, which plays important roles in the absorption and transport of β-carotene in H. cumingii. These results improve our understanding of the molecular mechanisms of nacre color formation in H. cumingii.

Modeling Post-Implantation Human Development to Yolk Sac Blood Emergence

Project description:Implantation of the human embryo commences a critical developmental stage that comprises profound events including axis formation, gastrulation, and the emergence of yolk sac hematopoietic system. Our mechanistic knowledge of this window of human life remains limited due to restricted access to in vivo samples for both technical and ethical reasons. Stem cell models of human embryo have emerged to help unlock the mysteries of this stage. Here, we present a model of early post-implantation human embryogenesis (embryoid) with an extra-embryonic endoderm and mesoderm niche to support early hematopoiesis. This human embryo model is genetically inducible, and its unanticipated self-organization results in the development of the amniotic cavity, formation of bilaminar disc embryonic morphology, an anterior hypoblast pole and posterior domain. The extra-embryonic layer of iDiscoid presents multilineage yolk sac tissue morphogenesis with extra-embryonic mesoderm and lacks trophoblast. Its development captures distinct waves of yolk sac hematopoiesis including the emergence of erythroid, myeloid and lymphoid cells . iDiscoid offers an easy-to-use, high-throughput, reproducible, and scalable platform to probe multifaceted aspects of human development and blood formation at early post-implantation stage. It will provide a tractable human-based model for drug testing, disease modeling, and a unique cell source for regenerative medicine.

A Spatiotemporal Transcriptome Revealed Stalk Development in Pearl Millet

Project description:Pearl millet is a major cereal crop that feeds more than 90 million people worldwide in arid and semi-arid regions. The stalk phenotypes of Poaceous grasses are critical for their productivity and stress tolerance, however, the molecular mechanisms governing stalk development in pearl millet remained to be deciphered. In this study, we spatiotemporally measured 19 transcriptomes for stalk internodes of four different early developmental stages. Data analysis of the transcriptomes defined 4 developmental zones on the stalks and identified 12 specific gene sets with specific expression patterns across the zones. Using weighted gene co-expression network analysis (WGCNA), we found that 2 co-expression modules together with candidate genes were involved in stalk elongation and thickening of pearl millet. Among the elongation-related candidate genes, we established by SELEX that a MYB-family transcription factor PMF7G02448 can bind to the promoters of three cell wall synthases genes (CesAs). In summary, these findings provide insights into stalk development and offer potential targets for future genetic improvement of pearl millet.
