Characterization of circulating PDGFRa positive cells in fetus

ABSTRACT: Mesenchymal stem/stromal cells (MSCs) are supplied to the damaged tissue via circulation when the injury is massive. MSCs recruited to the damaged tissues support tissue regeneration by differentiating into parenchyma cells and suppressing inflammation. However, most of the analysis of the circulating MSCs have been conducted in adult models, the existence and properties in fetal circulation was not known. Here, we have extensively analyzed the circulating MSCs cells during development.

Project description:Circulating osteoprogenitor (COP) are a population of cell in the peripheral circulation that possess functional and phenotypical characteristics of multipotent stromal cells (MSCs). While there is functional overlap, it is not known how COP cells are related to bone marrow (BM)-derived MSCs (BM-MSCs) and other better characterized stromal progenitor populations such as adipose-derived stromal cells (ASCs). This study compares COP cells to BM-MSCs and ASCs through detailed transcriptomic and proteomic analyses. COP cells have a distinct gene and protein expression pattern to BM-MSCs and ASCs, with a significantly stronger immune footprint, likely owing to their hematopoietic lineage. However, they also have a similar pattern of expression BM-MSCs and ASCs, in genes and proteins in progenitor cell differentiation and proliferation pathways. This study shows COP cells to be a unique but functionally similar population to BM-MSCs and ASCs, sharing their proliferation and differentiation capacity, but with a strong immune phenotype, with potential for translational regenerative medicine strategies.

Effects of light exposure on circulating metabolites in mice

Project description:The goal of this project is to identify metabolites released from subcutaneous adipose tissue due to light exposure that regulate systemic metabolism. We found that histidine levels increased by 8 days of blue light exposure, both in the treated subcutaneous adipose tissue and in circulation. Additionally, blue light-induced circulating histidine levels were not affected by the central blockade of histidine decarboxylase activity using FMH (Fluoromethylhistidine) in the hypothalamus. Circulating extracellular RNA markers of liver regeneration

Project description:Background and Aims: Although the liver can regenerate after sustaining injury, the tools to detect ongoing regeneration are lacking. The restoration of the liver after hepatectomy involves rapid, sequential, and coordinated changes in gene expression that result in systemic and local changes, such as the activation of progenitor cell populations and proliferation of quiescent hepatocytes. We postulated that changes in gene expression and regenerative programs could be activated within target cells through the transfer of extracellular RNA across cells or within the circulation, coordinating tissue response, and these extracellular RNA could represent biomarkers of the hepatic regenerative response. Methods: We identified temporal changes in circulating extracellular RNA at selected time periods up to 24 hours after partial hepatectomy in mice. (CD-1 males) Results: A peak increase in extracellular RNA content occurred 6 hours after hepatectomy. RNA sequencing was performed to identify the circulating extracellular non-coding RNA released after partial hepatectomy. To identify candidate markers, we used bioinformatics analyses to find the enriched RNA in the circulation after hepatectomy. We then performed tissue expression analysis for selected non-coding RNA and correlated it to these findings. Finally, a digital PCR assay was used to detect and quantify the expression of selected extracellular RNA in serum samples. Conclusions: Extracellular RNA that were selectively enriched during acute regeneration were detected within the circulation and represent biomarkers of ongoing liver regeneration in mice. The ability to detect ongoing active regeneration would benefit the evaluation of hepatic recovery after liver injury.

De novo sequencing of circulating microRNAs in locally advanced breast cancer

Discovery profiling of circulating small RNAs has not been previously reported in breast cancer (BC), and was carried out in this study to identify blood-based small RNA markers of BC clinical outcome. The pre-treatment sera of 42 stage II–III locally advanced and inflammatory BC patients who received neoadjuvant chemotherapy (NCT) followed by surgical tumor resection were analyzed for marker identification by deep sequencing all circulating small RNAs.

Genome-wide analysis of the circulating miRNome after cerebral ischemia reveals a reperfusion-induced microRNA cluster

Project description:Background and Purpose - Circulating microRNAs (miRNAs) are emerging biomarkers for stroke due to their high stability in the bloodstream and association with pathophysiologic conditions. However, the circulating whole-genome miRNAs (miRNome) has not been characterized comprehensively in the acute phase of stroke. Methods - We profiled the circulating miRNome in mouse models of acute ischemic and hemorrhagic stroke by next-generation sequencing (NGS). Stroke models were compared to sham-operated and naïve mice to identify deregulated circulating miRNAs. Top-ranked miRNAs were validated and further characterized by qRT-PCR. Results - We discovered 24 circulating miRNAs with an altered abundance in the circulation 3 hours following ischemia, whereas the circulating miRNome was not altered after intracerebral hemorrhage compared to sham-operated mice. Among the upregulated miRNA in ischemia, the top-listed miR-1264/1298/448 cluster was strongly dependent on reperfusion in different ischemia models. A time course experiment revealed that the miR-1264/1298/448 cluster peaked in the circulation around 3 hours after reperfusion and gradually decreased thereafter. Conclusions - Alteration of the miRNome in the circulation is associated with cerebral ischemia/reperfusion, but not hemorrhage, suggesting a potential to serve as biomarkers for reperfusion in the acute phase. The pathophysiological role of reperfusion-inducible miR-1264/1298/448 cluster, which is located on chromosome X within the introns of the serotonin receptor HTR2C, requires further investigation.

Circulating proinflammatory cytokines alter transcriptional activity within fibrotic tissue of Dupuytren’s disease patients

Project description:In this project, we have utilized blood and tissue samples from patients with Dupuytren’s disease (DD), a common fibroproliferative disorder of the hand, to identify increased circulating proinflammatory cytokines and assess alterations in the tissue using RNAseq and FTIR analysis. Our results indicate that specific cytokines in circulation play a direct role in local disease progression and may be useful for prospective identification of patients at risk for disease progression or recurrence as well as the development of novel therapeutic strategies. Furthermore, these results help to mechanistically link the well understood association of DD with chronic low-grade inflammatory states.
