CD248 knockdown in in vitro differentiated adipocytes exposed to hypoxia

ABSTRACT: Human adipose tissue derived stem cells were differentiated to adipocytes in vitro. At the end of differentiation, cells were treated with siRNA targeting CD248 followed by exposure to 1% oxygen levels. Microarray analysis were performed to identify differentially regulated genes.

Project description:Post-injury remodeling is a complex process involving temporal specific cellular interactions in the injured tissue where the resident fibroblasts play multiple roles, including inflammation induction and tissue remodeling. To dissect the molecular basis of these interactions, we performed single-cell and spatial transcriptome analysis in human and mouse hearts after myocardial infarction. A subset of fibroblasts identified by unique high expression of CD248 was strongly correlated with collagen synthesis and remodeling, and genetic deletion of CD248 in fibroblast ameliorated cardiac fibrosis and dysfunction following ischemia/reperfusion, highlighting the functional importance of CD248 positive fibroblasts in post-injury pathological remodeling. CD248 stabilizes TGF-βR Ⅰ protein and activates ACKR3 expression in fibroblast, leading to enhanced T cell adhesion and retention. This CD248 mediated fibroblast-T cell interaction is required to sustain fibroblast activation and scar expansion in injured heart. Disruption of this interaction using monoclonal antibody or Chimeric antigen receptor T cell reduces T cell infiltration, ameliorates ischemia/reperfusion-induced cardiac fibrosis and improves heart function. Therefore, single cell and spatial molecular profiling in post-injury heart has uncovered a CD248 specific subclass of fibroblast with a critical role in fibroblast-T cell interaction and a new potential therapy to treat tissue fibrosis.

Transcriptome analysis of human pancreatic preadipocytes and in vitro differentiated adipocytes

Project description:The aim of this study is to investigate the impact of the metabolic status on the transcriptome of isolated preadipocytes and in vitro differentiated adipocytes. We identified 38654 transcripts in pancreatic fat cells. We report that preadipocyte differentiation increased the abundance of mRNA levels of proteins related to adipogenesis and lipid metabolism. These changes in the transcriptome were absent or less pronounced in fat cells obtained from patients with prediabetes and type 2 diabetes. Meanwhile, mRNA levels of proteins involved in excellular matrix remodeling, angiogenesis and cytoskeleton organization were less abundant in adipocytes of all metabolic groups.

The tretment of siRNA was performed by electroporation one day before induction of differentiation in vitro. The cells were lyzed and RNA was purified on day 6 (mid differentiation) and day 13 (full differentiation) from differentiation start.

Short interfering RNAs (siRNAs)-based knockdown of OLFM2 or PKP2 in human adipocytes

Project description:We introduced into subcutaneous fully-differentiated human adipocytes from the same subject short interfering RNAs (siRNAs), thus enabling the study of functional consequences of OLFM2 (siOLFM2; n=3 biological replicates) and PKP2 (siPKP2; n=3) knockdown in these cells. A lipid-based transfection reagent (i.e. Lipofectamine 3000 reagent) was used to provide a transient (72h), convenient, reliable and efficient transfection into in vitro cultured and fully differentiated adipocytes. A non-targeting siRNA was used as control (siCTRL; n=4) for both modifications.
