PngCMs isolated exhibit better potential in cardiac repair and in CM subtype differentiation

ABSTRACT: CMs were isolated from Myh6-Cre/Esr1;R26-tdTomato mice at P1 and P7,which were permanently labeled with RFP upon treatment with tamoxifen immediately after birth, and injected resulting CMs into infarcted mouse hearts induced by LAD ligation. After two weeks, RFP+ cells were isolated by FACS and subjected to scRNA-Seq.

Project description:Aim: To determine RNA expression levels in the heart of mice 4 hours post Myc activation. Hearts were isolated from 15 day old wild-type (R26+/+) and R26CAG-c-MycERT2/+ mice following 4 hours of MycER activation via administration of (Z)-4-hydroxytamoxifen. Hearts were isolated from adult TetO-HRas; Myh6-tTA; R26CAG-c-MycERT2/+ and TetO-HRas; Myh6-tTA following 4 weeks of oncogenic H-Ras activation (Doxycycline withdrawal) and following 4 hours of MycER activation via administration of (Z)-4-hydroxytamoxifen.

RNA-seq of medullary thymic epithelial cell (mTEC) subsets in inducible Aire-lineage tracing mice

Project description:The goal of the study was to sequence mRNA expression from sorted medullary thymic epithelial cell (mTEC) subsets in inducible Aire-CreERT2.R26-Stopfl-tdTomato lineage tracing mice after a pulse chase. Four cell subsets were sorted 7 days after a single 2mg pulse of tamoxifen administered by oral gavage. 4 biological replicates (1,2,3,4) were collected derived from 12 pooled thymi per replicate. From the DAPI-;CD45-;EpCAM+ TEC pool, cells were sorted as: pre-Aire (MHCIIlo;RFP-), early-Aire (MHCIIhi;RFP-), late-Aire (MHCIIhi;RFP+), and post-Aire (MHCIIlo;RFP+). The data were used to identify differentially expressed genes across the four mTEC subsets to examine mTEC heterogeneity and identify novel mTEC subpopulations.

Enhanced engraftment, proliferation, and therapeutic potential in heart using optimized iPSC-derived cardiomyocytes

Project description:Human pluripotent stem cell-derived cardiomyocytes (CMs) are a promising tool for cardiac cell therapy. To optimize graft cells for cardiac reconstruction, we compared the engraftment efficiency of intramyocardially-injected undifferentiated-induced pluripotent stem cells (iPSCs), day4 mesodermal cells, and day8, day20, and day30 purified iPSC-CMs after initial differentiation by tracing the engraftment ratio (ER) using in vivo bioluminescence imaging. This analysis revealed the ER of day20 CMs was significantly higher compared to other cells. Transplantation of day20 CMs into the infarcted hearts of immunodeficient mice showed significant functional improvement. Moreover, the imaging signal and ratio of Ki67-positive CMs at 3 months post injection indicated engrafted CMs proliferated in the host heart. Although this graft growth reached a plateau at 3 months, histological analysis confirmed progressive maturation from 3 to 6 months. These results suggested that day20 CMs had very high engraftment, proliferation, and therapeutic potential in host mouse hearts. Transcription profiling of infarcted and non-infarcted left ventricles at different time points to study the molecular mechanism of myocardial infarction.

Project description:This dataset is a time series (1 hour [h], 4 hours, 24 hours, 48 hours, 1 week [w], and 8 weeks) intended to compare normal functioning left ventricles [lv + lv2] with infarcted [ilv] and non-infarcted left ventricles [nilv]. ilv samples are taken from the region between the LAD artery and the apex on a mouse with myocardial infarction. Lv2 samples are from the same region in a sham operated mouse. Nilv samples are taken from the region above the infartion and the left ventricle [lv] samples mimic that region in a sham mouse. Cardiomyocyte progenitors generated by direct reprogramming and molecular beacon selection attenuate ventricular remodelling after experimental myocardial infarction

Project description:Both naturally occurring and artificially engineered multipotent cardiac progenitor cells (CPCs) are associated with highly variable cardiomyocyte (CM) differentiation efficiencies, resulting in unpredictable and/or low CM yields. Therefore, CM-committed progenitors may be a more reliable cell product for therapeutic use. Here, we demonstrate that unipotent progenitors are part of the phenotype spectrum arising during direct reprogramming of neonatal cardiac fibroblasts towards CMs using lentiviral transduction of Gata4, Mef2c, Tbx5, and Myocd. Induced CM progenitors (iCMPs) simultaneously express CPC- and CM-specific proteins and can be selected via hairpin oligonucleotide molecular beacons targeting Myh6/7-mRNA–expressing cells. Purified iCMPs can be expanded extensively with preserved phenotype using ascorbic acid and give rise to CM-like cells in differentiation assays. When transplanted into infarcted mouse hearts, iCMPs prevent CM loss, attenuate fibrotic scarring, and preserve ventricular function. Given the clinical failure of conventional cardiac cell therapy using adult stem cells, further translational development of this iCMP approach is warranted.

Blockade of Wnt secretion by LGK974 after ischemia reperfusion injury

Project description:The LAD of C57BL6 mice were temporally occluded to induce ischemia-reperfusion injury of the heart. Mice received a daily dose of the porcupine inhibitor LGK974 starting directly after the surgery to prevent Wnt protein secretion. Control animals received a DMSO vehicle control. Hearts were extracted 48h after induction of ischemia-reperfusion injury. The infarcted area was identified and processed for RNAseq analysis.

Cardiac Myofibrillogenesis 1 is Spatiotemporally Modulated by the Molecular Chaperone UNC45B

Project description:Sarcomeres are fundamental to cardiac muscle contraction. Their impairment can elicit cardiomyopathies, leading causes of death worldwide. However, the molecular mechanism underlying sarcomere assembly remains obscure. We used human embryonic stem cell (hESC)-derived cardiomyocytes (CMs) to reveal step-wise spatiotemporal regulation of core cardiac myofibrillogenesis-associated proteins. We found that the molecular chaperone UNC45B is highly co-expressed with KINDLIN2 (KIND2), a marker of protocostameres, and later its distribution overlaps with that of muscle myosin MYH6. UNC45B-knockout CMs display essentially no contractility. Our phenotypic analyses further reveal that: 1) binding of Z-line anchor protein ACTN2 to protocostameres is perturbed due to impaired protocostamere formation, resulting in ACTN2 accumulation; 2) F-ACTIN polymerization is suppressed; and 3) MYH6 becomes degraded, so it cannot replace non-muscle myosin MYH10. Our mechanistic study demonstrates that UNC45B mediates protocostamere formation by regulating KIND2 expression. Thus, we show that UNC45B modulates cardiac myofibrillogenesis by interacting spatiotemporally with various proteins.

Expression dynamics of HAND1/2 in in vitro human cardiomyocyte differentiation

Project description:Hand1 and Hand2 are transcriptional factors, and knockout mice of these genes show left and right ventricular hypoplasia, respectively. However, their function and expression in human cardiogenesis are not well studied. To delineate their expressions and assess their functions in human cardiomyocytes (CMs) in vitro, we established two triple reporter human induced pluripotent stem cell lines, HAND1mCherry, HAND2EGFP, with MYH6-driven iRFP670 or constitutive tagBFP expression and investigated their expression dynamics during cardiac differentiation. On day 5 of the differentiation, HAND1 expression marked cardiac progenitor cells. We profiled the CM subpopulations on day 20 with RNA-sequencing and found that mCherry+ CMs showed higher proliferative ability than mCherry- CMs and identified CD105 as a surface marker of highly proliferative CMs. Finally, we revealed that LEF1 is a key regulator of proliferation and is repressively regulated by HAND1 and HAND2.
