Transcription profiling of Arabidopsis red 35S:PAP1 plants to investigate environmental regulation of leaf colour

Project description:Examine global gene expression patterns in control and 35S:PAP1 Arabidopsis plants upon environmental perturbation (light and temperature) over the course of the experiments. Experiment Overall Design: Red coloured 35S:PAP1, and empty vector control, plants of Arabidopsis thaliana Columbia were grown under room temperature, high light (RTHL, 22Â°C, 440 Âµmol m-2 s-1 irradiance) conditions to promote red leaf colouration. The growth conditions were then changed to a high day temperature, low light (HTLL: 30Â°C day, 150 Âµmol m-2 s-1 irradiance) stress treatment for six days, during which time the 35S:PAP1 leaves lost much of their red colouration, turning green. The growth conditions were then returned to a low temperature high light (LTHL, 15Â°C, 62% RH, 440 Âµmol m-2 s-1 irradiance) regime to restore the red colouration.

Transcriptome analyses of Pst DC3000-inoculated Arabidopsis thaliana Col-0 and 35S::CBP60g plants at normal and elevated temperature

Project description:We performed RNA sequencing of Pseudomonas syringae pv. tomato (Pst) DC3000-infected A. thaliana Col-0 and 35S::CBP60g plants at normal (23C) and elevated (28C) temperatures. 4-week-old plants were pre-incubated at 23C and 28C for 48h and then leaves were syringe-infiltrated with DC3000 bacterial inoculum. Plants were incubated at their respective temperatures (23C or 28C) for another 24h post-inoculation before tissue collection for RNA extraction. RNA samples for submitted for RNA sequencing and we found different clusters of DC3000-regulated genes that were similarly or differentially regulated between Col-0 and 35S::CBP60g at elevated temperature. Temperature-downregulated DC3000-induced genes in Col-0 plants that were restored in 35S::CBP60g plants were enriched for immunity/defense-related genes, including those essential for host salicylic acid (defense hormone) biosynthesis and accumulation.

Expression differences in wild type and 35S::CLE41 Arabidopsis stems

Project description:Arabidopsis CLE41 regulates vascular organisation, cell division and xylem differentiation. 35S::CLE41 lines are characterised by an increase in the number of undifferentiated cells in vascular bundles and by radial patterning defects. To identify further genetic components involved in controlling vascular development, we compared the transcriptomes of one 35S::CLE41 line with that of wild type counterparts using microarray analysis. Inflorescence stem tissue was compared in 5 weeks old plants 1 cm above the rosette leaves where vascular bundles of 35S::CLE41 plants contained an average of 100.7 ± 9.1 undifferentiated cells compared to 59.5 ± 5.5 in wild type counterparts. Genes with a putative role in vascular development demonstrate changes in gene expression between the two genotypes tested.
