Project description:The improvement of grain filling is the key issue for promoting wheat thousand grain weight and grain yield. The levels of polyamines (PAs) significantly affect grain filling in cereals, but the mechanism by which PAs affect grain filling in wheat is unclear. In the present study, six wheat cultivars whose grain filling differed were used, and their grain-filling characteristics and endogenous PA contents were measured. In addition, exogenous PAs were supplied during the wheat grain-filling period. The grain-filling characteristics, hormone levels, starch contents, and gene expression [based on RNA sequencing (RNA-seq)] in the grain were analyzed. The objective of the present study was to investigate the effects of PAs on grain filling in wheat. The results suggested that the direct synthetic pathway from putrescine (Put) to spermidine (Spd) in the grain was a key factor in promoting grain filling and thousand grain weight in wheat. Spd through regulates the grain-filling rate of inferior grain during the early grain-filling period to affecting the grain filling and thousand grain weight of wheat. The promotive effect of Spd on the grain filling of inferior wheat grain was notably related to carbohydrate metabolism in that grain. Spd significantly increased the zeatin (Z) + zeatin riboside (ZR) contents but reduced the ethylene (ETH) evolution rate in the inferior grain. In addition, Spd significantly increased the sucrose synthase (SS) and acid invertase (AI) activities in the inferior grain. These effects of Spd led to increased sucrose content in the inferior grain. These reasons might explain why Spd significantly promoted the filling and weight of inferior wheat grain.

Project description:The biological functions of differently expressed proteins between superior and inferior spikelet grains were investigated based on the isobaric tags for relative and absolute quantification to further clarify the mechanism of rice grain filling at the proteomic level, as well as the response of inferior spikelets to drought dress (-20 kPa or -40 kPa). Compared with superior spikelets, inferior ones had lower sink strength due to the lower sink activities (lower expressions of ADP-glucose pyrophosphorylase, granule-bound starch synthase, starch branching enzyme and pullulanase) and smaller sink sizes (lower abundances of structural proteins). The slower and later grain filling resulted from the weaker decomposition and conversion of photoassimilate and the slower cell division. Moderate drought stress (-20 kPa) promoted the grain filling of inferior spikelets through regulating the proteins associated with photoassimilate supply and conversion. These proteins may be important targets for rice breeding programs that raise the rice yield under drought condition. The findings offer new insights into rice grain-filling and provide theoretical evidences for better quality control and scientific improvement of super rice in practice.

Project description:Heat stress is one of the major abiotic stress factor that affects wheat yield. Especially, heat stress during grain filling affects grain yield besides reduced grain quality. So, in our present study, three genotypes with varied levels of tolerance to heat stress were chosen. They were subjected to heat stress at two stages for three days viz., early (11-14days-post-anthesis) and late (27-30dpa) grain filling independently under controlled conditions. At 14 and 30dpa, the spikes were harvested from control and stress conditions from all the three genotypes, grains were isolated and pulverized. Hence pulverized tissues are used for RNA extraction and further for transcriptome sequencing using HiSeq 4000. Data were analyzed to identify the genes involved in imparting heat stress tolerance.

In a climate change scenario, extreme weather event such as heatwaves will be more frequent especially during the grain-filling (GF) stage and could affect grain weight and quality of crops. Molecular mechanisms underlying the response to short heat stress (HS) have been widely reported for the hexaploid wheat (Triticum aestivum) but the regulatory heat stress mechanisms in tetraploid durum wheat (Triticum turgidum ssp. durum) remain partially understood. In this work, we performed a transcriptomic analysis of durum wheat grains to HS during early GF to identify key HS response genes and their predicted regulatory networks under glasshouse conditions.

Project description:Flag (FL) and second leaves (SL) in rice show differential aging patterns during monocarpic senescence. Coordination of aging programs in the top leaves is important for effective grain-filling. However, molecular bases for differential aging programs in the top leaves have not been systematically explored in rice. Here, we performed mRNA-sequencing of FL and SL at six time points during the grain-filling period. mRNA expression data revealed 6,365 genes showing aging-dependent expression changes in FL and/or SL. Of them, while 3047 genes showed shared aging-dependent expression patterns between FL and SL, 3058 genes showed differential expression patterns, which were classified into 5 major groups (G1-5) based on their differential expression patterns. Of the groups, G3 representing amino acid (AA) transport showed consistent differential age-dependent expression patterns in independent samples, whereas the other groups showed inconsistent differential expression patterns. Moreover, of AA transporters (AATs) in G3, long-distance AATs showed invariant differential age-dependent expression patterns after panicle removal, consistent to panicle removal-invariant differential nitrogen contents between FL and SL, known to be associated with protein concentration in grains. Our results suggest that long-distance AA transport is an invariant core transcriptional program of differential aging in rice top leaves for nitrogen remobilization during grain-filling.

Project description:Modern rice cultivars have large panicle, however, yield potential of these cultivars is often not fully achieved due to the poor grain-filling of their late-flowering inferior spikelets (IS). Our earlier work suggests broad transcriptional reprogramming during grain filling and shows a difference in gene expression between IS and the earlier-flowering superior spikelets (SS). However, the links between the abundance of transcripts and their corresponding proteins are unclear. In this study, a SWATH-MS (sequential window acquisition of all theoretical spectra-mass spectrometry) -based quantitative proteomic analysis has been applied between SS and IS. A total of 304 proteins, ranging from cellular components to biological processes, were observed to be differentially expressed between IS and SS. Detailed gene ontology analysis indicated that several biological processes including photosynthesis, protein metabolism, and energy metabolism are differentially regulated. Further correlation analysis revealed that abundance of most of the differentially expressed proteins is not correlated to the transcript levels, indicating an extra layer of gene regulation which may exist during rice grain filling. These findings raise an intriguing possibility that these candidate proteins may be crucial in determining the poor grain-filling of IS. Therefore, we hypothesize that the regulation of proteome changes not only occurs at the transcriptional, but also at the post-transcriptional level, during grain filling in rice.

Project description:The poor grain-filling initiation often causes the poor development of inferior spikelets (IS) which limits the yield potential of large panicle rice (Oryza sativa L.). However, it remains unclear why IS often has poor grain-filling initiation. In addressing this problem, this study conducted a field experiment involving two large panicle rice varieties, namely CJ03 and W1844, in way of removing the superior spikelets (SS) during flowering to force enough photosynthate transport to the IS. The results of this study showed that the grain-filling initiation of SS was much earlier than the IS in CJ03 and W1844, whereas the grain-filling initiation of IS in W1844 was evidently more promoted compared with the IS of CJ03 by removing spikelets. The poor sucrose-unloading ability, i.e., carbohydrates contents, the expression patterns of OsSUTs, and activity of CWI, were highly improved in IS of CJ03 and W1844 by removing spikelets. However, there was a significantly higher rise in the efficiency of sucrose to starch metabolism, i.e., the expression patterns of OsSUS4 and OsAGPL1 and activities of SuSase and AGPase, for IS of W1844 than that of CJ03. Removing spikelets also led to the changes in sugar signaling of T6P and SnRK1 level. These changes might be related to the regulation of sucrose to starch metabolism. The findings of this study suggested that poor sucrose-unloading ability delays the grain-filling initiation of IS. Nonetheless, the efficiency of sucrose to starch metabolism is also strongly linked with the grain-filling initiation of IS.
