Genome-wide maps of chromatin state in 5-HT6R knock out mice.

ABSTRACT: Purpose:The primary cilia play central roles in transducing or regulating various signaling pathways during brain development. However, the function of primary cilia in mature nerve system is currently largely unknown.We aimed to identify transcriptional changes resulting from depletion of ciliary GPCR 5-HT6R. Methods: We performed RNA sequencing in the hippocampal tissues from P90 adult konck out mice and wild type mice. Results:We examined for differential expression genes across 27698 genes expressed in our datasets. At significant cutoffs corresponding to Benjamini and Hochberg adjusted p value < 0.05, we found 429 differential expression genes in 5-HT6R-/- mice compared with WT litter mates. Conclusions:We discovered that primary cilia were associated with axon initial segment (AIS) morphology, including AIS length and position. Our data strongly suggest that the cilia signaling might have a remarked impact on neuronal excitability by regulating AIS morphology and are likely to be associated with neuronal system diseases.
