Project description:Abstract: Background: Hyposalivation is one of the common symptoms of diabetes. Although the micro ribonucleic acid (miRNA) has recently been identified to play a role in the pathogenesis of diabetes, the specific effects of miRNAs on diabetic salivary glands remain unclear. This work was aimed to investigate the expression profiles of miRNAs and identify miRNA‐mRNA network in submandibular gland (SMG) in db/db mice. Methods: miRNA and mRNA expressional profiles were analyzed by microarray technology from the SMG of db/db mice and db/m mice. Significant fold changes in miRNAs and mRNAs were confirmed by real-time quantitative reverse transcription-polymerase chain reaction. Bioinformatic analyses were performed to identify the critical biological functions and signaling pathways involved in diabetes-mediated hyposalivation and to identify the functional networks formed between differentially expressed (DE) miRNAs and mRNAs. Results: A total of 28 DE miRNAs and 1146 DE mRNAs were identified of the SMG in db/db mice and db/m mice. Gene ontology (GO) analysis and KEGG pathway analysis demonstrated that DE miRNAs were highly associated with terms related to diverse biological processes and signaling pathways. Of the related pathways, TGF-beta signaling pathway and tight junction pathway were notable. The constructed network indicated interactions between 11 miRNAs and 42 mRNAs. Autophagy pathway is worth studying, especially the dynamic network of miRNA and mRNA. In addition, AKT3 and PIK3CD may play important roles in the development of diabetes-mediated hyposalivation. Conclusions: Our research describes the miRNA and mRNA expression profiles and functional networks involved in SMGs from db/db mice. These results provide possible molecular mechanisms and may help to elucidate possible molecular therapeutic targets for the treatment of diabetes-induced hyposalivation. Project description:Hyposalivation is a complication of hypertension. However, little is known about the role of long non-coding RNAs (lncRNAs) in salivary glands in hypertension. This study aimed to compare the lncRNA and mRNA expression profiles between spontaneous hypertension rats (SHRs) and Wistar-Kyoto (WKY) rats through microarray analysis and apple bioinformatics methods to analyse their potential roles in hyposalivation. The differentially expressed (DE) lncRNAs and mRNAs were confirmed by quantitative real-time PCR (qRT-PCR). Compared with WKY rats, 225 DE lncRNAs and 473 DE mRNAs were identified in the SMG of SHRs. The pathway analyses of DE mRNAs showed that inflammatory mediator regulation of transient receptor potential channels was involved in hyposalivation in SHRs. Ten DE lncRNAs were chosen for further research. A coding-non-coding gene co-expression (CNC) network and competing endogenous RNA (ceRNA) network analysis revealed that the potential functions of these 10 DE lncRNAs were closely connected with the processes of the immune response. This study showed abundant DE lncRNAs and mRNAs in hypertensive SMGs. Furthermore, our results indicated strong associations between the immune response and hyposalivation and showed the potential of immune-related genes as novel and therapeutic targets for hyposalivation.

LncRNA expression profile of the SMGs was analyzed using microarray technology. Differentially expressed lncRNAs were confirmed using real-time quantitative PCR. Bioinformatics analyses were performed, and Coding-non-coding gene co-expression (CNC) and competing endogenous RNA (ceRNA) networks were constructed to explore the potential mechanisms of diabetes-induced hyposalivation.ResultsA total of 1273 differentially expressed lncRNAs (536 up-regulated and 737 downregulated) were identified in the SMGs tissues of db/db mice. CNC and ceRNA network analyses were performed based on five differentially expressed lncRNAs validated by real-time quantitative PCR. Gene Ontology analysis of target genes of CNC network revealed that "calcium ion binding" was a highly enriched molecular function. Kyoto Encyclopedia of Genes and Genomes pathway analysis of target genes of ceRNA network revealed that the "mammalian target of rapamycin signaling pathway" was significantly enriched.ConclusionsOn the whole, the findings of the present study may provide insight into the possible mechanism of diabetes-induced hyposalivation.

Project description:Inflammation and its consequent fibrosis are two main features of diabetic nephropathy (DN), but target therapy on these processes for DN remains yet ineffective. We report here that miR-29b is a novel therapeutic agent capable of inhibiting progressive renal inflammation and fibrosis in type 2 diabetes in db/db mice. Under diabetic conditions, miR-29b was largely downregulated in response to advanced glycation end (AGE) product, which was associated with upregulation of collagen matrix in mesangial cells via the transforming growth factor-? (TGF-?)/Smad3-dependent mechanism. These pathological changes were reversed by overexpressing miR-29b, but enhanced by knocking-down miR-29b. Similarly, loss of renal miR-29b was associated with progressive diabetic kidney injury, including microalbuminuria, renal fibrosis, and inflammation. Restored renal miR-29b by the ultrasound-based gene therapy was capable of attenuating diabetic kidney disease. Further studies revealed that inhibition of Sp1 expression, TGF-?/Smad3-dependent renal fibrosis, NF-?B-driven renal inflammation, and T-bet/Th1-mediated immune response may be mechanisms associated with miR-29b treatment in db/db mice. In conclusion, miR-29b may play a protective role in diabetic kidney disease and may have therapeutic potential for diabetic kidney complication.
