RNAseq of old and young males with equal physical activity

ABSTRACT: 18 older (65-71 years) and 20 young (19-25 years) males were recruited based on similar, self-reported physical activity, which was verified using accelerometry measurements. Gene expression profiles of vastus lateralis biopsies obtained by RNA sequencing. A clear ageing signature was observed despite similar physcical activity.

Project description:RNA-sequencing from 12 young (20-35 years old) and 12 older (60-85 years old) participants' skeletal muscle and adipose tissue. Biopsies were taken at rest following an overnight fast, and following alcohol/caffeine restriction for 24 hours, and no strenuous physical activity for 48 hours. Skeletal muscle was taken by the Bergstrom technique under local anaesthesia (Lidocaine hydrochloride) from the dominant leg Vastus Lateralis, and adipose tissue was taken by needle aspiration 5cm lateral to the umbilicus.

Exercise Training Effects on Human Skeletal Muscle DNA Methylation

Project description:This study aimed to determine skeletal muscle DNA methylation changes in a cohort of volunteers with a range of insulin sensitivities following 8-weeks of supervised exercise training. We studied 13 sedentary participants (5M/8F, 34.6 ± 3.1 years) and performed euglycemic hyperinsulinemic clamps with vastus lateralis muscle biopsies and peak aerobic activity (VO2 peak) tests before and after training. We extracted DNA from the muscle biopsies and performed global methylation using Illumina's Methylation EPIC 850K BeadChip.
