ILC2 promote type 2 immunity in colonic epithelial cells during N. brasiliensis infection.

Project description:ILC2 contribute to tissue immunity by secreting large amounts of cytokines upon activation. However, their specific contribution to worm infection and epithelial remodeling remains incompletely understood. We tested the impact of ILC2 on tissue immunity in the colon of mice infected with the nematode Nippostronylus brasiliensis. We performed scRNA sequencing of colon epithelial cells from mice specifically lacking ILC2 (Nmur1-Cre Id2 fl/fl). Transcripts typically associated with type 2 immunity were reduced in ILC2-deficient mice, indicating a non-redundant role of ILC2 for type 2 tissue immunity in worm protection that cannot be compensated by other cell types. These findings highlight the importance of ILC2 for nematode control and type 2 tissue immunity.

Beta-naphthoflavone treated worms

Project description:The soil nematode Caenorhabditis elegans is one of the simplest animals having the status of a laboratory model. Its genome contains 80 cytochrome P450 genes (CYP). In order to study CYP gene expression in C. elegans mixed stages and synchronized hermaphrodites were exposed to 18 known xenobiotic cytochrome P450 inducers. Messenger RNA expression was detected by DNA arrays and semiquantitative RT-PCR. Using subfamily-specific primers, a pooled set of exon-rich CYP fragments could be amplified. In this way it was possible to systematically check the influence of different inducers on CYP expression at the same time. The well-known CYP1A inducers beta-naphthoflavone, PCB52, and lansoprazol were the most active and in particular they strongly induced almost all CYP35 isoforms. A few number of further CYP forms were found to be inducible by other xenobiotics like phenobarbital, atrazine, and clofibrate. In addition, a transgenic C. elegans line expressing GFP under control of the CYP35A2 promoter showed a strong induction of the fusion by beta-naphthoflavone in the intestine. RNA-seq analysis of Arabidopsis thaliana wild-type roots and type-A arr3,4,5,6,7,8,9,15 mutant roots non-infected and infected with Heterodera schachtii nematodes

Project description:Plant-parasitic cyst nematodes induce the formation of hypermetabolic feeding sites, termed syncytia, as their sole source of nutrients. The formation of the syncytium is orchestrated by the nematode in part by modulation of phytohormone responses, including cytokinin. In response to infection by the nematode H. schachtii, cytokinin signaling is transiently induced at the site of infection and in the developing syncytium. Arabidopsis lines with reduced cytokinin sensitivity show reduced susceptibility to nematode infection, indicating that cytokinin signaling is required for optimal nematode development. Furthermore, lines with increased cytokinin sensitivity also exhibit reduced nematode susceptibility. To ascertain why cytokinin hypersensitivity reduces nematode parasitism, we examined the transcriptomes in wild-type and a cytokinin-hypersensitive type-A arr Arabidopsis mutant in response to H. schachtii infection. Genes involved in the response to biotic stress and defense response were elevated in the type-A arr mutant in the absence of nematodes and were hyper-induced following H. schachtii infection, which suggests that the Arabidopsis type-A arr mutants impede nematode development because they are primed to respond to pathogen infection. These results suggest that cytokinin signaling is required for optimal H. schachtii parasitism of Arabidopsis, but that elevated cytokinin signaling triggers a heightened immune response to nematode infection.

Transcriptome analysis in the germline of wild-type (WT) n2 worm and nmad-1(ok3133) mutant at 20°C and 25°C.

Project description:The goal of RNA-seq is to identify the differential expressed genes in the wild-type worm and nmad-1 mutant worm at 20°C and 25°C respectively. 100 pairs of germlines were extracted and collected from worm tips. Two biological replicates were assigned for each group and the RNA profiles were generated by deep sequencing, using NEXTseq 500 Illumina. We mapped about 20 - 30 million reads per sample to C. elegans genome (build ce10) with bowtie2 workflow. Differential expression genes were identified through EBSeq in RSEM pipeline with 0.05 p value and 1.2 fold change. Our results showed elevated number of misregulated genes in the nmad-1 mutant groups at 25°C.
