Project description:The aim of this study was to identify differential gene expression resulting from the inhibition of RORgt in human CD4+ T cells. Human CD4+ T cells were cultured with anti-CD3/anti-CD28 mAb for 20 to 60 hours concurrent with exposure to Vehicle (DMSO), TMP778 (0.2 uM) or TMP776 (0.2 uM). All samples were then processed for RNA extraction, labeling, and hybridization to Affymetrix arrays.

Project description:Mouse spleen CD4+CD44LowCD62LHigh cells were stimulated with 2ug/ml anti-CD3(eBioscience), 2ug/ml anti-CD28 (eBioscience) in the presence (Th17 condition) or absence (Th0 condition) of hTGFb-1 (1ng/ml eBioscience), mouse IL-6(10ng/ml eBioscience). Cells were collected after 24 hourÕs culture using RNAeasy columns (Qiagen) for total RNA isolation. Isolated total RNA from Th0 and Th17 conditioned cells were submitted for the microarray gene transcription comparison analysis using Affymetrix Mouse 430A 2.0 array chips.

To unravel the role of Ikaros in the expression of pro-inflammatory genes without Th17 polarizing cytokines, we performed an RNA-seq analysis in naive CD4+ T cells and CD4+ T cells activated with anti-CD3+anti-CD28 antibodies (Th0 condition), from WT (Ikaros f/f CD4-Cre-) and Ikaros TKO mice (Ikaros f/f CD4-Cre+).

Effect of acute pyrvinium pamoate treatment on gene expression of CD4+ T cells during early activation.

Project description:Treatment of T cells with pyrvinium pamoate (PP) early during their activation blocks mitochondrial biogenesis and results in reduced proliferation, skewed CD4+ T cell differentiation and reduced cytokine production. Therefore, to examine the role of PP treatment on mitochondria health, we performed gene expression analysis to identify differentially expressed genes in naive CD4+ T cells cultured with anti-CD3/CD28 plus PP or DMSO. Naive CD4+ T cells were activated for six hours with 2 mg/mL anti-CD3/anti-CD28 mAbs and activated for an additional one hour in the presence or absence of PP.
