Genetic and pharmacological targeting of hepatic oxalate overproduction ameliorates metabolic dysfunction-associated steatohepatitis

Project description:Metabolic dysfunction-associated steatohepatitis (MASH) is on the rise, and with limited pharmacological therapy available, identification of new metabolic targets is urgently needed. Oxalate is a terminal metabolite produced from glyoxylate by lactate dehydrogenase (LDHA). The liver-specific alanine-glyoxylate aminotransferase (AGXT) detoxifies glyoxylate, preventing oxalate accumulation. We report that AGXT is suppressed and LDHA is activated in livers from patients and mice with MASH, leading to oxalate overproduction. In turn, oxalate promotes steatosis in hepatocytes by inhibiting peroxisome proliferator activated receptor-alpha (PPARα) transcription and fatty acid β-oxidation (FAO), and induces monocyte chemotaxis via C-C motif chemokine ligand 2. In male mice with diet-induced MASH, blocking oxalate overproduction through hepatocyte-specific AGXT overexpression or pharmacological inhibition of LDHA potently lower steatosis, inflammation, and fibrosis by inducing PPARα-driven FAO, and suppressing monocyte chemotaxis, nuclear factor-kappaB and transforming growth factor-beta targets. These findings highlight hepatic oxalate overproduction as a new target for the treatment of MASH.

Expression factors for oxalate stimulation in HEK293T cells

Project description:parental HEK293T were challenged with 1064 µg/cm2 calcium oxalate monohydrate (COM) or sodium oxalate (NaOx) 4mM or vehicle for 24 hours and DNA microarray was performed. We observed and selected two-fold upregulation of laminin, beta 3 (LAMB3), early growth response 1 (EGR1), gremlin 1, DAN family BMP antagonist, Ca++-dependent secretion activator 2, Ras association domain family member 3, interleukin 33 and bone morphogenetic protein 8a , and two-fold down-regulation of interleukin 37 and intercellular adhesion molecule 1 in COM lord compared to vhicle.

Analyse the differently expressed genes in tamoxifen resistant breast cancer cells

Project description:Tamoxifen resistance has been a major clinical problem and is accountable for relapse in about one third of ER positive breast cancer patients. Most of the recurrent patients will eventually receive chemotherapy. However, the chemosensitivity of these tamoxifen-resistant breast cancer patients has never been explored. In this study, we demonstrate that tamoxifen-resistant breast cancer cells express significantly more BARD1 and BRCA1, which results in the resistance to DNA-damaging chemotherapy including cisplatin and adriamycin , but not to paclitaxel. Silencing BARD1 or BRCA1 expression or inhibition of BRCA1 phosphorylation by Dinaciclib restored the sensitivity to cisplatin in tamoxifen-resistant cells. In addition, we identified that activated PI3K/AKT pathway in tamoxifen-resistant cells was responsible for the upregulation of BARD1 and BRCA1. PI3K inhibitors, BKM120 and BYL719, decreased the expression of BARD1 and BRCA1 in tamoxifen-resistant cells and re-sensitized them to cisplatin both in vitro and in xenografted mice. Higher BARD1 and BRCA1 expression was associated with poor prognosis of early breast cancer patients, especially the ones received radiotherapy, indicating the potential use of PI3K inhibitors to reverse chemoresistance and radioresistance in ER positive breast cancer patients.

Comparative transcriptomics analysis of monocytes of heathy human subjects before and after a dietary oxalate load

Project description:Next gen RNA Seq analysis was performed to study the effect of oxalate load on the transcriptome of monocytes in healthy human subjects. Healthy subjects consumed a fruit and vegetable smoothie containing high amount of oxalate. Blood was collected from them before and 5 hrs after consumption of the smoothie. Monocytes were purified from the blood and RNA Sequencing was performed by Illumina NextSeq 500 platform. Differential gene expression analysis was performed using DESeq2 considering fold change > + 2.0 with a q value of < 0.05. 1197 genes were found to be differentially expressed which were used for pathway and functional analysis using Ingenuity software.
