Identification of hub genes and key miRNAs associated with oxidative stress in radiation-induced lung injury via RNA sequencing and bioinformatics analysis


ABSTRACT: In this study, through X-ray irradiation and RNA sequencing of human bronchial epithelial BEAS-2B cells, central genes and enrichment pathways related to oxidative stress of radiation lung injury were found, and the potential molecular mechanism of RILI was elucidated.The GO function annotation and KEGG pathway enrichment analysis of candidate genes were performed using David database.The STRING database and Cytoscape platform were used to construct PPI network diagram.The result was 421 DEGs.The candidate genes (158 up-regulated genes and 101 down-regulated genes) enriched 289 GO terms and 38 KEGG pathways.The top 10 pivot genes and the top 3 key miRNAs were found by constructing the network map.In summary, we identified central genes, key miRNAs and enrichment pathways related to oxidative stress, as well as the potential molecular mechanisms of RILI.These results provide new ideas for elucidating the potential molecular mechanism of RILI and identifying new targets and new approaches for prevention and control of RILI.
