RNA-seq of Human Endometrial B Cells During the Mid-luteal Phase

ABSTRACT: We present the transcriptome analysis by RNA sequencing of endometrial B cells identifying potential B cell phenotypes and function in human endometrium during the mid-luteal phase. To achieve this, endometrial B cells were isolated from 15 endometrial biopsies, and cDNA library preparation was performed for RNA sequencing.

Biomarker discovery in paired mid luteal endometrial biopsies

Project description:Cell state determination is apparent within the endometrium during the mid-luteal window of implantation. Using previously identified biomarkers of cell states (SCARA5 and DIO2) we performed bulk RNA sequencing on 6 pairs of endometrial biopsies characterised by either normal biomarker expression in both biopsies, or abnormal biomarker expression in one biopsy (low SCARA5 and high DIO2) and normal expression in a second biopsy. We demonstrate that “abnormal cycles” are associated with a distinct gene expression profile when compared to “normal cycles” and have utilised this dataset for biomarker discovery.

Single-cell RNA sequencing of human endometrium across the luteal phase of the menstrual cycle

Project description:The fate of the human endometrium is determined during the mid-luteal window of implantation, a crucial period when endometrial stromal cells (EnSCs) differentiate into specialized decidual cells. Upon embryo implantation, these differentiating EnSCs transform the endometrium into the decidua of pregnancy. Conversely, in the absence of pregnancy, decreasing progesterone levels trigger tissue breakdown, leading to menstruation. Despite our understanding of these processes, the precise mechanisms governing this tissue transformation remain elusive. To bridge this knowledge gap, we conducted single-cell RNA sequencing, mapping the transcriptomic profiles of timed endometrial biopsies throughout the luteal phase of the menstrual cycle.

Project description:<p>We performed an unbiased genome-wide expression quantitative trait locus (eQTL) mapping study to identify common regulatory (expression) single nucleotide polymorphisms (eSNPs) in mid-secretory endometrium, corresponding to the luteal phase of the ovarian cycle. Biopsies were collected from 58 women with two or more early pregnancy losses and 53 of these samples passed all quality control. RNA was extracted from endometrial biopsies and DNA was extracted for sequencing from blood.</p> <p>Gene expression data can be found at NCBI GEO Series GSE77688.</p>

Project description:In order to try and identify characteristics of gene expression in the endometrium of women suffering infertility or recurrenty miscarriage, we performed RNAseq on endometrial pipelle biopsies from 20 women. The endometrial transcriptome in the mid-luteal phase of the cycle (window of implantation) is highly divergent in women suffering infertility or miscarriages. 20 mid-luteal endometrial biopsies were analysed from infertile women and patients suffering recurrent pregnancy loss.
