Functional genomics study of oral squamous cell carcinoma under PPARG overexpression by RNA-seq

ABSTRACT: Oral squamous cell carcinoma (OSCC), one of the most common malignant tumor in head and neck, which is regulated by cellular factors. Among them, peroxisome proliferator activated receptor gamma (PPARG) has been reported to have main functions in the regulation of several human cancer cells, including OSCC. However, its regulatory mechanism remains unclear. Here, we performed a functional genomics study of PPARG overexpressing OSCC by using RNA-seq.

Glutamine synthetase regulates the alternative metabolism of γ-aminobutyrate in oral squamous cell carcinoma

Project description:More than 90% of cancer cases in head and neck region are oral squamous cell carcinoma. Recent studies identified several tumor-specific metabolites for the screening or diagnosis of OSCC patients. However, the metabolic reprogramming of OSCC is not well understood. Here, we compared the metabolites between cancerous and paracancerous tissues of OSCC patients and investigated the metabolism of γ-aminobutyrate in OSCC derived cells. Our data revealed that the increase of γ-aminobutyrate was promoted by the synthesis of glutamate beyond the mitochondria, which was regulated by glutamine synthetase. This study is not only benefit for understanding the pathological mechanisms of OSCC, but also has application prospects for the diagnosis and therapy of OSCC.

Obesity Potentiates TH2 Immunopathology Via Dysregulation of PPARg

Project description:Clinically, obesity is strongly associated with severe TH2 immunopathology, though the physiological, cellular, and molecular underpinnings of this association remain obscure. We demonstrate that obese mice are susceptible to severe atopic dermatitis (AD), a major manifestation of TH2 immunopathology and disease burden in humans. Mechanistically, we show that dysregulation of the nuclear hormone receptor peroxisome proliferator-activated receptor gamma (PPARg) in T cells is a causal link between obesity and the increased TH2 immunopathology. We find that PPARg directly controls a cellular metabolic transcriptional program that restrains nuclear gene expression of the chief TH2 effector cytokines interleukin-4 (IL-4) and interleukin-13 (IL-13). Accordingly, thiazolidinediones (TZDs), potent PPARg agonists, robustly protect obese mice from TH2 immunopathology. Collectively, these findings establish PPARg as a molecular link between obesity and TH2 immune homeostasis and identify TZDs as novel therapeutic candidates for TH2 immunopathology.

RBAP48 modulates the oral squamous cell carcinoma process through AR-dependent and independent mechanisms

Project description:Oral squamous cell carcinoma is a head and neck malignancy induced by multiple factors. The progression of OSCC involves epithelial cell proliferation from fibrosis to malignancy. However, the underlying molecular mechanisms that influence these processes remain unclear. Here, we identify RBAP48, an important AR co-activator involved in OSCC cancer progression. RBAP48 is highly expressed in OSCC tissues and has a poor prognosis. As a DHT-dependent AR coactivator, RBAP48 not only up-regulates the transcriptional activity of AR but also affects the transcription of target genes in the cell cycle and external stimulation pathways throughout the genome. RBAP48 can interact with SP1, AR, and other transcription factors in OSCC cells and co-recruit the promoter region of CCND1 and RAB31. Finally, we find that RBAP48 can promote the growth of OSCC cells, stimulate the response to drugs and external substances in vitro and in vivo, and ultimately promote cancer progression. These results suggest that RBAP48 may be an effective target for predicting the progression and treatment of OSCC cancer.

Disruption of PPARg signaling results in mouse prostatic intraepithelial neoplasia (mPIN) involving active autophagy

Project description:Peroxisome proliferator-activated receptor-gamma (PPARg) regulates the interface between cellular lipid metabolism, redox status and organelle differentiation. Following conditional prostatic epithelial knockout of PPARg in mice we observed focal hyperplasia of the epithelium which developed to mouse prostatic intraepithelial neoplasia (mPIN), becoming progressively more severe with time. We selectively knocked down PPARg2 isoform in wild-type mouse prostatic epithelial cells and examined the consequences of this in a tissue recombination model. Histopathologically the results resembled the conditional PPARg KO mouse prostates. Electron microscopy showed accumulated defective lysosomes and autophagic vacuoles in both of PPARg- and g2- deficient cells. Gene expression profiling indicated a major dysregulation of cell cycle control and metabolic signaling networks related to peroxisomal and lysosomal maturation, lipid oxidation and degradation. We conclude that PPARg maintains the maturation and turnover of peroxisomes and lysosomes in prostate epithelium. Disruption of PPARg signaling results in autophagy and oxidative stress during mPIN pathogenesis. The mPrE-PPARg knockout and mPrE-PPARg2 shRNA cells were compared to wildtype mPrE cells. Time (3 days culture) and cell types (x 4) were tested.

DNA methylation analysis of oral squamous cell carcinoma

Project description:Upper aerodigestive tract (UADT) tumors present different biological behavior and prognosis, suggesting specific molecular mechanisms underlying their development. However, they are rarely considered as single entities (particularly head and neck subsites) and share the most common genetic alterations. Therefore, there is a need for a better understanding of the global DNA methylation differences among UADT tumors. We performed a genome-wide DNA methylation analysis of esophageal (ESCC), laryngeal (LSCC), oral (OSCC) and oropharyngeal (OPSCC) squamous cell carcinomas, and their non-tumor counterparts. The unsupervised analysis showed that non-tumor tissues present markedly distinct DNA methylation profiles, while tumors are highly heterogeneous. Hypomethylation was more frequent in LSCC and OPSCC, while ESCC and OSCC presented mostly hypermethylation, with the latter showing a CpG island overrepresentation. Differentially methylated regions affected genes in 127 signaling pathways, with only 3.1% of these being common among different tumor subsites, but with different genes affected. The WNT signaling pathway, known to be dysregulated in different epithelial tumors, is a frequent hit for DNA methylation and gene expression alterations in ESCC and OPSCC, but mostly for genetic alterations in LSCC and OSCC. UADT tumor subsites present differences in genome-wide methylation regarding their profile, intensity, genomic regions and signaling pathways affected.
