Analysis of differentiatng monocytes transcriptome changes in response to type I IFNs

ABSTRACT: To analyze the transcriptome changes of differentiating monocytes in response to type I IFNs, RNA seq was performed. The MACS-purified mouse BM monocytes were cultured in the presence or absence of I IFNs for 12 h or 24h under the condition of M-CSF, samples were harvested for total RNAs and subjected to RNA sequencing analyses.

Project description:I IFNs promote differentiating monocytes to differentiate into tumor-associated macrophages (TAMs) and limit the I IFNs anti-tumor effects.But how macrophage polarization is shaped by complex signals in the tumor microenvironment during IFN therapies is incompletely understood. Herein, it was discovered that I IFNs could stimulate transitional monocytes to secrete interleukins IL-4 and IL-6, which interact to influence target genes expression in primary mouse bone-marrow-derived macrophages (BMDMs). To analyze the synergism mechanism, RNA seq was performed. Bone marrow derived macrophages were cultured under the 20ng/ml M-CSF for 7days, and treated with the indicated cytokine, samples were harvested for total RNAs and subjected to RNA sequencing analyses.

Glycoproteomes of naive and LPS stimulated human monocytes and THP-1 cells

Project description:CD14+ Monocytes from healthy volunteers were purified by MACS (negative selection) and FACSorting and either left untreated or stimulated for 24h and 48h with LPS. THP-1 cells were stimulated for 4h, 24h and 48h with LPS. Glycoproteins were captured with hydrazide chemistry and tryptic and PNGase F-released peptide fractions analyzed by MS/MS. Quantitative assessment revealed differential glycoprotein expression in activated/LPS-tolerized monocytes and naïve monocytes and THP-1 cells.

Project description:Analysis of genes induced in DC precursors and in BM cells and monocytes treated with GM-CSF For progenitor arrays, bone marrow progenitors (CMP, GMP, CDP, and pre-cDC) were harvested from WT C57Bl/6 mice. For culture arrays, BM was cultured in the presence of GM-CSF or M-CSF and adherent and non-adherent cells sorted. Project description:Comparison of the DNA methylation profiles of CD14+ monocytes from human peripheral blood with derived dendritic cells (DCs) and macrophages (MACs) obtained by exposure with GM-CSF/IL-4 and GM-CSF, respectively, and with mature DCs and MACs after lipopolysaccharide (LPS) exposure The methylation profiles of bisulfite-modified DNA of human CD14+ monocytes were compared with derived immature dendritic cells (iDCs) and macrophages (iMACs) following GM-CSF/IL-4 and GM-CSF incubation, and then activation/maturation with lypopolysaccharyde (LPS) using the Infinium HumanMethylation450 BeadChips (Illumina, Inc., San Diego, CA,). This platform allows the interrogation of >485,000 methylation sites per sample at single-nucleotide resolution, and comprises an average of 17 CpG sites per gene in the 99% of RefSeq genes. 96% of CpG islands are covered, with additional coverage in CpG island shores and the regions flanking them. The samples were hybridized in the array following the manufacturerâs instructions. Total DNA isolated by standard procedures from CD14+ cells (total monocytes, MOs) corresponding to three sets of samples of monocytes (MOs), derived immature DCs and MACs (iDCS and iMACS) and activated/mature DCs and MACs following incubation with LPS (mDCS and mMACs)

Human Monocytes to M-CSF differentiated Macrophages

Project description:This dataset was created to study M-CSF dependent in vitro differentiation of human monocytes to macrophages as a model process to demonstrate that independent component analysis (ICA) is a useful tool to support and extend knowledge-based strategies and to identify complex regulatory networks or novel regulatory candidate genes. Keywords: M-CSF, Monocytes, Macrophages Samples for microarray analysis were isolated from healthy donors and from donors with Niemann-Pick type C disease. Monocytes were differentiated to macrophages for 4 days in the presence of M-CSF (50 ng/ml,R\&D Systems). Differentiation was confirmed by phase contrast microscopy. Total RNA was extracted from the tissue biopsies according to the manufacturerM-BM-4s instructions using the RNeasy Protect Midi Kit (Qiagen). Purity and integrity of the RNA was assessed on the Agilent 2100 bioanalyzer with the RNA 6000 Nano LabChip reagent set (Agilent Technologies, BM-CM-6blingen, Germany). The RNA was quantified spectrophotometrically and then stored at M-bM-^HM-^R80M-BM-0C. Gene expression profiles were determined using Affymetrix HG-U133A and HG-U133plus2.0. Array scanning and expression analysis was performed using Microarray Analysis Suite 5.0 software.
