Gene expression analysis of murine alveolar macrophages in the presence of lung cancer cells

ABSTRACT: To investigate the interaction between lung alveolar macrophages and lung cancer cells in vivo, gene expression analysis was performed using orthotopic tumor bearing animal model with C57BL/6 mice and Lewis Lung Carcinoma (LLC) cells. CD45+, F4/80+, Siglec-F+ population was sorted as alveolar macrophage population with fluorescence-activated cell sorting (FACS) technique.

Cancer-cell transcriptional signatures associated with Socs1 knockdown in an immunocompetent and orthotopic mouse model of non-small lung cancer

Project description:How cancer cells respond to signals coming from the tumor microenvironment cannot be fully elucidated in vitro. Additionally, RNA-Sequencing data from bulk human tumors does not assess the contribution of cancer cells alone, but from a complex mixture of both host and cancer cells. Using an immunocompetent and orthotopic mouse model of human Non-small Cell Lung Cancer, we sought to determine cancer cell specific transcriptomes from bulk tumors and compared these to cancer cells grown in vitro. We hypothesized that knockdown of Socs1, or suppressor of cytokine signaling 1 in Lewis Lung Carcinoma (LLC) cells would not only impact their response to single-agent immunotherapy, but also lead to complex changes in the tumor microenvironment. First, we harvested RNA from either LLC-NT (LLC cells transduced with a non-targeting control vector) or LLC-sh21 (LLC cells transduced with a Socs1 knockdown vector) cell lines grown in vitro. To determine the transcriptome of these cells grown in vivo, we injected LLC-NT or LLC-sh21 cells into the left lung of transgenic GFP-expressing C57BL/6J mice. After three weeks, tumor-bearing lung lobes were isolated from mice and were made into single cell suspensions. The GFP-negative cancer cell population was sorted from GFP-positive host cells by fluorescence activated cell sorting. RNA was extracted from freshly isolated cancer cells grown in vitro or from cancer cells isolated from tumors. Our RNA-seq analysis shows that LLC-sh21 cells respond to IFNg coming from the microenvironment relative to LLC-NT cells which have a blunted response due to high basal expression of SOCS1. These data suggest that specific genes in the IFNg response pathway are important for response to immunotherapy.

To determine genes regulated by TDAG8 in vitro, mouse Lewis lung carcinoma (LLC) cells stably expressing TDAG8 (TDAG8-LLC cells) and control vector (control LLC cells) were cultured under acidic conditions. Transcriptome microarray analysis using Affymetrix GeneChip Mouse Genome 430 2.0 Arrays was performed with total RNA prepared from these cells. To determine genes regulated by TDAG8 in vitro, mouse Lewis lung carcinoma (LLC) cells stably expressing TDAG8 (TDAG8-LLC cells) and control vector (control LLC cells) were cultured under acidic conditions. Transcriptome microarray analysis using Affymetrix GeneChip Mouse Genome 430 2.0 Arrays was performed with a mixture of equal amounts of total RNA samples from four independent cell cultures.

Gene expression signatures for murine alveolar type II lung epithelial cells after tumor bearing

Project description:To determine the influence of primary tumors on pre-metastatic lungs, we have employed whole genome microarray expression profiling as a discovery platform to identify gene signatures of alveolar type II epithelial cells (AT-II) in TLR3 deficient mice (Tlr3-/-) and wide-type (WT) littermates with tumor bearing. We subcutaneously inoculated Tlr3-/- and WT mice with Lewis lung carcinoma (LLC). Two weeks later, lung tissues from Tlr3-/- and WT mice were dissociated and AT-II cells were sorted. AT-II cells from mice without tumor bearing were set as controls.

Characterization of tumor microenvironment using single cell RNA sequencing in Lewis Lung Carcinoma cell transplantation mouse model

Project description:The tumor microenvironment holds significant importance in cancer development and progression. Research conducted using mouse models of tumor cell transplantation can enhance our comprehension of the tissue microenvironment that allows tumor growth at an early stage. Among these models, those featuring the Lewis Lung Carcinoma (LLC) cell line are noteworthy due to their low immunogenic features. Since numerous cancer patients do not respond effectively to current immunotherapy, it is imperative to investigate models with low immunogenic features. Consequently, this study employed single-cell RNA sequencing on an orthotopic lung cancer mouse model utilizing the LLC cell line to profile the characteristics of diverse cell types that emerge in the lung during tumor engraftment. The lung microenvironment exhibited discernible transformations, including fibrogenic gene expression, increased cell-to-cell and cell-to-extracellular matrix adhesion, and reduced infiltration of T cells compared to lungs without tumor engraftment. In conclusion, these findings reveal cellular and molecular alterations that can be targeted for tumor prevention and treatment for low-immunogenic tumors.
