Project description:We performed a hybridization-based microarray analysis for lncRNA expression to understand the involvement of lncRNAs in HCC and to investigate differentially expressed lncRNAs in HCC, thereby characterizing their potential roles in tumor growth. The expression profiles of SMMC7721 HepG2 and Huh7 human HCC cell lines and the HL-7702 human immortalized normal hepatocyte cells were performed by the lncRNA microassay. SMMC7721, HepG2 and Huh7 human HCC cell lines were defined as the experimental group and HL-7702 cell line was used as the control.

T Regulatory Lymphocytes (Treg) Depletion for Cancer Treatment Efficacy and Safety Study

Project description:T regulatory lymphocytes were shown to be partly responsible for immune tolerance to cancer cells. In that respect these cells oppose to the mounting of an efficacious immune response needed to cure cancer. To treat advanced metastatic colorectal cancer, the investigators propose an immunotherapy consisting in autologous lymphocytes infusion depleted from T-regulatory cells, associated with a 5-day prior lymphoid-ablative chemotherapy associating cyclophosphamide (day 1 & 2) with fludarabine (day 1 to 5). To administer treatment and monitor chemotherapy safety, patients will be hospitalized for 3 weeks until complete recovery from chemotherapy. Patients will then be followed-up ambulatory for 9 months during which time they will be assessed for tumor size with computed tomography (CT) - scan (primary criteria).

Project description:To determine the physiological roles of NCOA4 and NCOA4-mediated ferritinophagy in hippocampal neurons, NCOA4 was depleted using siRNA and desferrioxamine (DFO; 100 µM, 24 h) was added to restrict iron availability. Cells were treated for NCOA4 depletion, iron restriction, or both, and then the transcriptome profiles acquired from RNA-seq were compared with those of control (scramble siRNA-treated) cells.

S-Adenosylmethionine Depletion Activates La Ribonucleoprotein Domain Family Member 1 to Induce Protein Translation

Project description:S-adenosylmethionine (SAMe) is the principal methyl donor synthesized by methionine adenosyltransferase 1A (MAT1A)-encoded enzyme in the liver. Mice lacking Mat1a have hepatic SAMe depletion, spontaneous development of non-alcoholic steatohepatitis (NASH) and hepatocellular carcinoma (HCC). To understand how SAMe depletion drives liver pathologies we performed phospho-proteomics in Mat1a knockout (KO) mice livers and the most striking change was hyperphosphorylation of La-Related Protein 1 (LARP1), which in the unphosphorylated form negatively regulates translation of 5'-terminal oligopyrimidine (TOP)-containing mRNAs. Consistently, multiple TOP proteins are induced in the KO livers. We identified LARP1-T449 as a novel, SAMe-sensitive phospho-site of cyclin-dependent kinase 2. LARP1-T449 phosphorylation induced global translation, cell growth, migration, invasion, and expression of oncogenic TOP-ribosomal proteins in HCC cells. LARP1 expression is increased in human NASH and HCC. Our results reveal a novel SAMe-sensitive mechanism of LARP1 phosphorylation that may be involved in the progression of NASH to HCC.

Project description:Hepatocellular carcinoma (HCC) is a cancer with global impact and largely refractory to current treatments. Novel treatment options are therefore urgently needed. LncRNAs play important regulatory roles in HCCs and are emerging as promising therapeutic options against HCC. We identified TP53TG1 as a promising tumor suppressor LncRNA. To understand TP53TG1 target genes and the mechanisms, we performed mRNA expression profiling in TP53TG1-overexpressing Huh7 cells and negative control.

Gene expression of human hepatocellular carcinoma cells in response to acyclic retinoid

Project description:To better understand the molecular basis of the anticancer effects of acyclic retinoid (ACR), a genome-wide screening was applied to identify novel targets of ACR in human hepatocellular carcinoma (HCC) cells JHH7. Gene expression profiles of JHH7 were measured at 0h, 1h and 4 hours after treatment with1 μM All-trans retinoic acid (AtRA) or 10 μM ACR. Hierarchical clustering with Ward’s method of 44,907 genes demonstrated diverse expression changes in HCC cells treated with ACR for 4h. A total of 973 differentially expressed genes in response to ACR by comparing with AtRA for 4h treatments were identified with a fold change more than 2. Then, network analysis was performed on the altered gene expression profiles using Ingenuity Pathways Analysis (IPA) program. The most highly populated networks were associated with the regulation of cell cycle and DNA replication, as ACR is well known to induce apoptosis and suppress cell proliferation in HCC cells. Moreover, networks related with amino acid metabolism, protein synthesis and lipid metabolism, such as the biological network entitled “Lipid Metabolism, Small Molecular Biochemistry, Vitamin and Mineral Metabolism” were also observed. Of interest, this network contains genes that play critical roles in controlling the development of tissues and organs such as the nuclear orphan receptor nuclear receptor subfamily 2, group F, member 2 (NR2F2), suggesting potential drug targets to prevent/treat HCC. Gene expression profiles of JHH7 were measured at 0h, 1h and 4 hours after treatment with 1μM AtRA or 10 μM ACR.
