Blast wave effect on brain proteome (rat hippocampus)

Project description:These data show differences in up- and down-regulation for protein abundances in the hippocampus of double blast vs. sham rats. Tandem mass tags (TMT)-MS results showed 136 up-regulated and 94 down-regulated proteins between the two groups. These TMT-MS findings revealed changes never described before in blast studies. In the absence of behavioral changes, these proteomic data further support the existence of an asymptomatic blast-induced molecular altered status (ABIMAS) associated with specific protein changes in the rat hippocampus.

Project description:Male Long Evans rats aging 10-12 weeks were exposed to repeated blast overpressure and then went through behavioral tests for anxiety and fear conditioning. After 1-1.5 months (sub-acute) or 12-13 months (chronic), amygdala tissue specimens from all animals were collected. RNA-seq was used to measure transcriptome-wide gene expression of sham and blast animals at different timepoints. The study identified changes in the amygdalar transcriptome and associated anxiety-related phenotypic outcomes related to both blast exposure and aging, which may play a role in the long-term pathological consequences of mTBI.

Analysis of gene expression in mouse brain regions after exposure to pulse-modulated 1.9 GHz radiofrequency fields

Project description:This study assessed changes in gene expression in 7 mouse brain regions using Illumina Mouse WG-6 (v2) beadchips after exposure to 1.9 GHz pulse-modulated radiofrequency field exposure for 4h/day for 5 days. High dose RF field exposure was whole body average SAR and brain average SAR were 1.45 W/kg and 0.16 W/kg, respectively. Total RNA isolated from the amygdala, auditory cortex, caudate, cerebellum, hippocampus, hypothalamus and medial prefrontal cortex of 1.9 GHz pulse-modulated radiofrequency field exposed mice and compared to that in sham (handled) exposed animals. An unhandled control group was also included in the study design. A total of 20 RNA samples (5 independent biological tissues for each of 4 treatment groups) were collected for each brain region. When analysis of gene expression was conducted within individual brain regions when controlling the false discovery rate (FDR), no differentially expressed genes were identified relative to the sham control. However, it must be noted that most fold changes among groups were observed to be less than 1.5 fold and this study had limited ability to detect such small changes. While some genes were differentially expressed without correction for multiple-comparisons testing, no consistent pattern of response was observed among different RF-exposure levels or among different RF-modulations. The current study provides the most comprehensive analysis of potential gene expression changes in the rodent brain in response to RF field exposure conducted to date. Within the exposure conditions and limitations of this study, no convincing evidence of consistent changes in gene expression was found in response to 1.9 GHz pulse-modulated RF field exposure.

Transcription profiling of rat cerebellum and hippocampus following exposure to neurotoxicant Aroclor 1254

Project description:Pregnant rats (Long-Evans) were dosed perinatally with 0 or 6Â mg/kg/day of Aroclor 1254 from gestation day 6 through postnatal day (PND) 21. Gene expression in cerebellum and hippocampus from PND7 and PND14 animals was analyzed with an emphasis on developmental aspects. Changes in gene expression (>= 1.5 fold) in control animals identified normal developmental changes. These basal levels of expression were compared to data from Aroclor 1254-treated animals to determine the impact of gestational PCB exposure on developmental parameters. The results indicate that the expression of a number of developmental genes related to cell cycle, synaptic function, cell maintenance, and neurogenesis is significantly altered from PND7 to PND14. Aroclor 1254 treatment appears to dampen the overall growth-related gene expression levels in both regions with the effect being more pronounced in the cerebellum. Functional analysis suggests that Aroclor 1254 delays maturation of the developing nervous system, with the consequences dependent on the ontological state of the brain area and the functional role of the individual gene.
