Single cell RNA-seq of Nutlin3a-Induced Senescent mouse embryonic fibroblasts with or without expressing OSKM

ABSTRACT: Purpose: Use scRNA-seq to determine the trajectories of transcriptomes during partial reprogramming of nutlin-3a senescent MEFs. Methods: MEFs were isolated from p16-CreERT2/rtTA3-mKate2/TRE-OSKM-mCherry mice. After the senescence induction through RO3306 and nutlin-3a, treatments with 4-OHT for 9 days and doxycycline for 12 days were performed to induce OSKM expression. The doxycycline treated MEFs and untreated MEFs were collected by trypsinization and directly used for scRNA library preparation through droplet system.

Project description:This study has examined the molecular mechanisms underlying sensitivity of sarcomas to Nutlin-3a, a non-genotoxic activator of the p53 pathway. Human patient material was collected immediately following surgical resection, dissected into small pieces and ex planted onto gelatin sponges immersed in media containing either vehicle control or Nutlin-3a (10uM and/or 50uM) for 48 hours. Nutlin-3a represents a novel targeted therapy for the treatment of sarcomas.
