MiRNA-seq of lungs tissue of swine supplemented with cholecalciferol and calcidiol

ABSTRACT: Results indicate that both increasing the dose of cholcalciferol and the use of calcidiol in the diet have a significant effect on lung tissue miRNAs compared to using the standard dose of cholecalciferol. The greatest changes in the transcriptome were induced by supplementation with the increased dose of the combination of cholecalciferol and calcidiol.

Project description:Maintaining an appropriate concentration of vitamin D is essential for the proper functioning of the body regardless of age. Nowadays, there are more and more indications that vitamin D supplementation at higher than standard doses may show protective and therapeutic effects. Our study identifies differences in the body's response to long-term supplementation of cholecalciferol at an increased dose. Two groups of pigs were used in the experiment. The first group received a standard dose of cholecalciferol (grower- 2000 and finisher- 1500 IU/kg feed), and the second group received an increased dose (grower- 3000 and finisher- 2500 IU/kg feed). The animals were slaughtered when they reached breeding maturity. Then the lung tissue was collected from the animals. The material was used for mRNA sequencing. The results of mRNA sequencing showed a significant change in the expression of 195 genes. Functional analysis using the KEGG database showed that increased cholecalciferol supplementation significantly activated 42 pathways. Most of the activated pathways is associated with metabolism.

Microbiome overview in swine lungs.

Project description:Mycoplasma hyopneumoniae is the etiologic agent of swine enzootic pneumonia. However other mycoplasma species and secondary bacteria are found as inhabitants of the swine respiratory tract, which can be also related to disease. In the present study we have performed a total DNA metagenomic analysis from the lungs of pigs kept in a field condition, with suggestive signals of enzootic pneumonia and without any infection signals to evaluate the bacteria variability of the lungs microbiota. Libraries from metagenomic DNA were prepared and sequenced using total DNA shotgun metagenomic pyrosequencing. The metagenomic distribution showed a great abundance of bacteria. The most common microbial families identified from pneumonic swine's lungs were Mycoplasmataceae, Flavobacteriaceae and Pasteurellaceae, whereas in the carrier swine's lungs the most common families were Mycoplasmataceae, Bradyrhizobiaceae and Flavobacteriaceae. Analysis of community composition in both samples confirmed the high prevalence of M. hyopneumoniae. Moreover, the carrier lungs had more diverse family population, which should be related to the lungs normal flora. In summary, we provide a wide view of the bacterial population from lungs with signals of enzootic pneumonia and lungs without signals of enzootic pneumonia in a field situation. These bacteria patterns provide information that may be important for the establishment of disease control measures and to give insights for further studies.
