Urine Large Extracellular Vesicle LC-MS/MS Data from a Chronic Kidney Disease Rat Model

Project description:This study examined large renal tubule extracellular vesicles (LRT-EVs) isolated from urine of a 5/6 nephrectomy (5/6Nx) rat model that recapitulates many elements of human chronic kidney disease (CKD). Within weeks of 5/6Nx rats spontaneously exhibit proximal tubular damage including brush boarder shedding and the production of LRT-EVs. Analysis of the LRT-EV size and a lack of TUNEL staining in 5/6Nx rats, suggested LRT-EVs to be distinct from exosomes, microvesicles, and apoptotic bodies.

Acidosis activates the Nrf2-pathway in renal proximal tubule-derived cells through a crosstalk with renal fibroblasts

Project description:Crosstalk of renal epithelial cells with interstitial fibroblasts plays an important role in kidney pathophysiology. A former study showed that crosstalk between renal epithelial cells and renal fibroblasts protects against acidosis-induced damage. In order to gain further mechanistic insight into this crosstalk we investigated the effect of acidosis on the transcriptome of renal epithelial cells (NRK-52E) and renal fibroblasts (NRK-49F) in co-culture by RNASeq, bioinformatics analysis and experimental validation.

Male-dominant activation of rat renal organic anion transporter 1 (Oat1) and 3 (Oat3) expression by transcription factor BCL6

Project description:Organic anion transporters 1 (Oat1) and 3 (Oat3) play a critical role in transport of organic anions, including frequently prescribed drugs, across cell membranes in proximal tubules of kidneys. In rats, these transporters are known to be male-predominant and testosterone dependently expressed. The molecular mechanisms that are involved in the sex-dependent expression are unknown. Our aim was to identify genes that show a sex-dependent expression and could be involved in sex-dependent regulation of Oat1 and Oat3. Comparison of gene expression in cortical kidney slices between 4 male and 4 female RCCHanTMWIST rats. Each animal were analyzed as a separate sample.
