Three control MRTFA-/-;MRTF-BFlox/Flox and three MRTF-A-/-;MRTF-BF/F;GFAP-Cre RNA samples were collected and pooled for hybridization on Affymetrix microarrays.

Project description:Neuronal Elavl-like (nElavl) RNA binding proteins have three paralogs (Elavl2, Elavl3 and Elavl4) in the mouse genome. This family of RNABPs have been implicated in a variety of post-transcriptional RNA processing mechanisms, including regulation of mRNA stability, alternative splicing, and translational regulation. In this study, using mouse exon arrays, we identify significant differences (p<0.01) in 119 transcripts between wild-type and Elavl3/Elavl4 double knockout forebrain tissue at postnatal day 0.

Mouse retina from Msi1/Msi2 double knockout in photoreceptor cells

Project description:This quantitative protepomics study (TMT10 isobaric labeling) of the protein expression in the retina of Msi1/Msi2 double knockout mouse compared to floxed controls. The Msi1 and Msi2 genes were knocked out in photoreceptor cells using tamoxifen inducible Cre (Cre-ERT2) under the control of Pde6g promoter. Tamoxifen was administered by intrapertoneal injection for three consecutive day starting at postnatald day 30, and the retina was collected at postnatal day 51. Retinas from floxed animals lacking the Cre recombinase and treated with tamoxifen were used as controls. Five biological replicates for each knockout (three femailes, two males) and control (two femaes and three males) group were used.

Project description:During the first 10 postnatal days of mouse development, Purkinje cells, neurons of the cerebellum, pass through different developmental processes of differentiation such as profound dendritic remodeling and growth, cell death and loss of their ability to regenerate their axons. To identify the genes involved in these different processes and in particular transcription factors, the gene expression patterns of murine cerebellar cortical area centered on Purkinje soma have been measured using Affymetrix microarrays at 5 different developmental stages (postnatal days P0, P3, P5, P7 and P10; 4 replicates = 4 independent measurements for each stage).
