Single-nucleus RNA-seq of mouse arcuate nucleus of the hypothalamus after DOCA-salt treatment

ABSTRACT: We compared the transcriptomic changes in cell populations of the arcuate nucleus of the hypothalamus in 21 day treated DOCA-salt male mice versus sham male mice using high-throughput single-nucleus RNA-seq.

Project description:m6A, the most prevalent internal modification on mRNAs, plays important roles in the nervous system. Whether neurogenesis in the hypothalamus, a region critical for controlling appetite, is regulated by m6A signaling, especially in humans, remains unclear. Here we showed that deletion of m6A writer Mettl14 in the mouse embryonic hypothalamus led to adult obesity, with impaired glucose-insulin homoeostasis and increased energy intake. Mechanistically, deletion of Mettl14 leads to hypothalamic arcuate nucleus neurogenesis deficits with reduced generation of feeding-related neurons and dysregulation of neurogenesis-related m6A-tagged transcripts. Deletion of m6A writer Mettl3, or m6A reader Ythdc1, shared similar phenotypes. METTL14 or YTHDC1 knockdown also led to reduced generation of feeding-related neurons in human brain subregion-specific arcuate nucleus organoids. Our studies reveal a conserved role of m6A signaling in arcuate nucleus neurogenesis in mice and human organoids and shed light on the developmental basis of epitranscriptomic regulation of food intake and energy homeostasis.

Single-nucleus transcriptome sequencing reveals a specific neuronal subtype in the arcuate nucleus.

Project description:Over recent decades, research has established the hypothalamus, particularly the arcuate nucleus, as a central hub for the homeostatic control of energy balance. The hypothalamic arcuate nucleus is a pivotal regulator of energy balance, containing anorexic proopiomelanocortin (POMC) neurons and orexigenic agouti-related peptide (AgRP) neurons, which exert opposing effects on energy homeostasis. However, the molecular identity of “non-AgRP, non-POMC” neurons remains unclear. We identified a distinct neuronal subtype characterized by the absence of Agrp and Pomc expressions, while exhibiting robust Crabp1 expression.

Arcuate nucleus of the hypothalamus cells are sensitive to nutritional state in a sex dependent manner.

Project description:Obesity has reached epidemic levels and obesity increases the risk for negative health outcomes including type 2 diabetes. Sex differences are evident in metabolic homeostasis with differences seen both in amount and location of adipose stores. Males store more adipose viscerally whereas females store adipose subcutaneously. Interestingly, this can be affected by the brain. The arcuate nucleus of the hypothalamus is a key brain region that exerts control over feeding behavior and metabolism. The cells of the arcuate are heterogeneous in their transcriptomic expression and in their functions. Here we perform single nuclei RNA-sequencing of the arcuate in females and males under varying nutritional states. We found that Agrp neurons were the most sensitive to nutritional state and this was more pronounced in females than in males. KNDy neurons were also sensitive to nutritional state and major sex differences were observed. Dopamine neurons of the arcuate had major sex differences. Ghrh neurons saw moderate nutritional regulation and sexual dimorphism. Both microglia and oligodendrocytes had moderate differences observed between nutritional states. This study represents the first major effort to characterize sex differences across nutritional states in the arcuate.

Analyses of Agrp neuron transcriptomes reveals differential expression of receptors for multiple neuromodulators. Neuroanatomical mapping of known ligands of the receptors define subsets of neurons directly upstream of AgRP neurons in specific brain areas.

Project description:Coupling the release of pituitary hormones to the developmental stage of the oocyte is essential for female fertility. It requires estrogen to restrain kisspeptin (Kiss1) neuron pulsatility in the arcuate hypothalamic nucleus, while also exerting a surge-like effect on Kiss1 neuron activity in the AVPV hypothalamic nucleus. However, a mechanistic basis for this region-specific effect has remained elusive. Here, we provide functional insight into hypothalamic estrogen sensing by analyzing estrogen receptor alpha (ERα/ESR1) DNA binding events in the arcuate and AVPV nuclei of the hypothalamus in female mice.

The transcriptomics of SLC18A3 positive cells from arcuate nucleus of rats in different pubertal development stages.

Project description:Based on our spatial transcriptomics sequencing data, we observed solute carrier family 18 member A3 (SLC18A3) was specificially expressed in kisspeptin-expressed neurons in arcuate nucleus (ARC). SLC18A3 as a surface marker was used to examine the isolation of living kisspeptin-expressed neurons by flow cytometric (FACS) assay. Fresh isolated ARC nucleus tissues from female Sprague-Dawley rats in postnatal day (PND)-25, 35 and 45 (n=10 each group) were harvested, and processed single-cell suspension for FACS assay by SLC18A3. Here, we obtained two populations according to the cell size by FSC/SSC signals, and further harvested a small proportion of Slc18a3+ cells from hypothalamus of PND-25, 35 and 45. Smart-seq was conducted to detect the transcriptome of four population namely large Slc18a3+/- cells as well as small Slc18a3+/- cells.
