The role of GCNT1 mediated O-glycosylation in aggressive prostate cancer.

ABSTRACT: To identify signalling pathways altered by GCNT1 in prostate cancer cells, we established PC3 stable cell lines in which GCNT1 was overexpressed. We then performed differential gene expression, Gene Ontology and GSEA analyses using data obtained from RNA-seq of the control and OE cell lines.

MEK5-mediated gene expression changes in PC3 human prostate cancer cells

Project description:Mitogen/extracellular signal-regulated kinase kinase-5 (MEK5) belongs to the family of MAP kinases. It is activated by the upstream kinases MEKK2 and MEKK3. MEK5, in turn, phosphorylates and activates extracellular signal-regulated kinase 5 (ERK5) at Thr218/Tyr220. MEK5/ERK5 pathway plays a pivotal role in tumor initiation and progression, including prostate cancer. MEK5 protein is overexpressed in prostate cancer cells compared with normal cells and MEK5 levels are correlated with prostate cancer metastasis. Global gene expression was determined in PC3 cells stably expressing a scrambled (control) shRNA or MEK5 shRNA using the Agilent Mouse Whole Genome microarrays. Gene ontology and pathway analysis of differentially expressed genes using Ingenuity Pathway Analysis and gene set enrichment analysis (GSEA) revealed MEK5 knockdown attenuates a number or critical signaling pathways required for prostate tumor growth and progression.

Phosphoproteomics analysis to identify PPP2R5D substrates

Project description:The purpose of this project is to identify the substrates that are specific for B'delta regulatory subunit. For this purpose, we have generated two doxycycline-inducible stable cell lines (OE and RE) that express B'delta. In the OE cell lines, B'delta is overexpressed without downregulation of the endogenous regulatory subunits. On the other hand, in the RE cell lines, B'delta is overexpressed concurrently with a downregulation of the endogenous regulatory subunits. To stimulate protein phosphorylation, we used isoproterenol, a beta-adrenergic agonist that is known to induce the PKA-mediated phosphorylation and activation of the B'd.

Effect of NO-sulindac treatment on hypoxic prostate cancer cells

Project description:The hypoxia response contributes to radio and chemo-resistance in cancer cells. Our previous work has shown that the nitric oxide donating non-steroidal anti-inflammatory drug (NO-NSAID) NO-sulindac is a potent inhibitor of the hypoxia response in prostate cancer cells and leads to increased susceptibilty to radiation. In this study we used microarrays to investigated the global impact of NO-sulindac on the hypoxia response in prostate cancer cells with a view to determining the mechanism of action. PC3 hormone-insensitive prostate cancer cells were grown under normoxic or hypoxic conditions and treated with NO-sulindac, unnitrated sulindac or vehicle control. Global gene expression in response to treatment was examined using microarrays and the bioconductor software suite. Gene set enrichment analysis (GSEA), Gene ontology (GO) analysis and pathway analysis were used to examine the biological impact of treatments.

Therapeutic targeting of splicing in aggressive prostate cancer

Project description:Androgen deprivation therapy (ADT) is the main therapeutic regimen for patients with advanced prostate cancer (PCa). However, most treated patients invariably develop the castration-resistant disease (i.e., CRPC). Mechanisms underlying CRPC development and maintenance remain poorly understood. Recent studies have established splicing dysregulation as a new molecular hallmark of cancer. However, the functional and clinical relevance of such misregulation have not been systematically explored in PCa. Experimentally, we show that splicing modulator, E7107, can significantly inhibits CRPC cell growth both in vitro and in vivo. To determine the mechanistic origins of therapeutic effect of E7107 in vitro, we treated AR+ LNCaP and AR- PC3 cells with DMSO or E7107 at 10nM for 6 h, followed by deep RNA-seq analysis of rRNA-depleted total RNAs isolated from biological triplicates. Furthermore, to evaluate the activity of E7107 against CRPC in vivo, we utilized Enzalutamide-resistant LAPC9-AI (androgen-independent) and PC3 models. RNA-seq experiments we performed on tumors exactly 4 h after the fifth injection of E7107 (5mg/kg/day, 5-days injection considered as 1-dose treatment). Five tumors were included for each group (vehicle or E7107) for deep RNA-seq.

RNA-Seq analysis of prostate cancer cells after morsuin treatment

Project description:We performed RNA-seq analysis using different prostate cancer cell lines (PC3 and 22Rv1) after morusin treatment. We reported that morusin regulates Akt/mTOR pathway-associated genes in PC3 and 22Rv1 cells. Morusin induces apoptosis and suppresses cell cycle progression, cell migration and invasion in PC3 and 22Rv1 cells.
