EWS-FLI1-transfection of HEK293

Project description:Response of HEK293-cells after transfection with EWS-FLI1. HEK293 cells were transfected with the expression vector pIRES2-EGFP containing type I EWS-FLI1 or empty control vector. For transient transfection standard DEAE dextran method was used and RNA was isolated 48h post transfection. For stable transfection cells were transfected using FuGENE 6 (Roche, Mannheim, Germany) and cells were selected with 400 ug/mL G418. Transient transfection of miR-449 family in the hepatocellular carcinoma cell line HLE

Project description:In this study, we comparatively analyzed the members of the miR-449 family (miR-449a, miR-449b, and miR-449c) with regards to their target genes and functional effects in hepatocellular carcinoma (HCC). Microarray analysis after transient transfection of miR-449a, miR-449b, and/or miR-449c in the HCC cell line HLE identified putative target genes of miR-449a, miR-449b, and miR-449c. For transient overexpression of miRNAs, the HCC cell line HLE was transfected with 50 nM Allstars Negative Control or miScript miRNA mimics. MiRNA mimics for miR-449a, miR-449b, and miR-449c were either single transfected or cotransfected in equimolar amounts (16.7 nM each). Global mRNA expression profiling was performed utilizing pooled RNA of three biological replicates per microarray and two microarrays per condition.

Effect of 4-Hydroxynonenal on gene expression. Comparison of HSF1-siRNA silenced vs control cells

Project description:4-Hydroxynonenal (HNE), a cytotoxic and diffusible electrophile generated by the spontaneous decomposition of oxidized lipids, has a suspected role in neurodegenerative and inflammatory disease processes. In addition to promoting cell death, elevated levels of HNE lead to the engagement of cytoprotective signaling pathways, including the heat shock, antioxidant, DNA damage, and ER stress responses. Activation of the heat shock response, mediated by the transcription factor heat shock factor 1 (HSF1), is critical for maintaining cellular viability in the presence of HNE. Accordingly, silencing HSF1 expression using siRNA enhances the toxicity of HNE. Microarray analysis of samples from control and HSF1-silenced cells was performed to investigate which associated changes in gene could be responsible for the decrease in cellular viability. Four different experimental conditions were investigated. Samples were prepared and analyzed in triplicate. Samples 1-3 received NEG control siRNA and DMSO (vehicle) for 6 h. Samples 4-6 received NEG control siRNA and 50 uM HNE for 6 h. Samples 7-9 received HSF1 siRNA and DMSO (vehicle) for 6 h. Samples 10-12 received HSF1 siRNA and 50 uM HNE for 6 h.

Conditional knock-out of Transferrin receptor 1 in Alb-cre H11-myc mice

Project description:Disrupted iron metabolism is commonly observed in various types of cancer. However, the role of iron signaling in controlling the tumor microenvironment and its clinical relevance remain unclear. We found that intra-tumoral iron signals stabilized PD-L1 protein, dampening CD8+ T cell responses and promoting tumor evasion. Blocking iron uptake decreased PD-L1 expression and enhanced CD8+ T cell infiltration, leading to attenuated tumor growth. Mechanistically, iron preserved PD-L1 protein integrity by inhibiting its ubiquitination. Iron-induced lipid peroxidation in tumors promoted the generation of 4-Hydroxynonenal (4-HNE), which subsequently adducted to the PD-L1 cytoplasmic domain for its carbonylation. 4-HNE-mediated carbonylation outcompeted PD-L1 ubiquitination, resulting in PD-L1 protein stabilization. Finally, we introduced a designed peptide in tumor cells to diminish PD-L1 carbonylation by competing for 4-HNE availability. This led to decreased PD-L1 expression and enhanced CD8+ T cell immunity, hindering tumor growth. Importantly, such peptide therapy showed effective outcomes in anti-PD-L1 non-responding tumors. Thus, our findings reveal alternative strategies for overcoming PD-L1-mediated immune evasion in cancer.
