Effect of MetAP2 the inhibitor SDX-7320 on gene expression in BT-474 xenografts

ABSTRACT: Analysis of gene expression using bulk RNASeq was performed on tumors dissected from vehicle-treated and SDX-7320-treated mice (12 mg/kg). The analysis of RNASeq data showed that a several pathways were modulated in tumors from mice treated with SDX-7320.

RNAseq analysis of wildtype and Nrf2-/- advanced lung tumors

Project description:Nrf2 plays a critical role in lung cancer. We investigated the gene expression profile in Nrf2 wildtype and knockout tumors. Nrf2 wildtype and knockout mice were injected with 2 or 4 doses of vinyl carbamate (16 mg/kg). Lung tumors were then dissected and pooled from each group to isolate total RNA. RNAseq was then performed to analyze the differential expression profiles between groups.

Effects of BT-11 treatment on hippocampal gene expression in Female Fisher transgenic 344-AD rats. [Female RNAseq]

Project description:5 month old Female Fisher transgenic 344-AD (Tg-AD) rats expressing human Swedish amyloid precursor protein (APPsw) and Δ exon 9 presenelin-1 (PS1ΔE9) were treated with BT-11 (cat. no. HY-102013, MCE) at 8.5 mg/kg body weight administered in rodent chow. Following 6 months of BT-11 treatment, rats (11 months of age) were tested for cognitive behavior prior to sacrificing. Hippocampal Brain region were dissected and RNA sequencing (RNAseq) analyses was done on the Isolated sample. Taken together we showed that BT-11 treatment enriched pathways for females included passive transmembrane transporter activity, G protein-coupled receptor activity, G protein-coupled peptide receptor activity.

Lasting response by vertical inhibition with Cetuximab and Trametinib in KRAS mutated Colorectal Cancer Patient-derived Xenografts (PDX)

Project description:Global mRNA expression profiling of patient derived colon cancer xenograft models (N=13) were collected using Agilent human whole genome array (G4845A AMADID 026652, cRNA 4x44k V2) . All xenograft models carried an activating. All Mice were either untreated (controls) or treated with treated with cetuximab (25 mg/kg, twice weekly, i.p., Merck) and trametinib (0.5 mg/kg, five subsequent days per week, p.o., Hycultec) to establish response characteristics. Tumors showing disease control under the combination treatment were chronically treated to establish cetuximab secondary resistant tumors which was successful for BoC2 and BoC56 .Primary resistant PDX tumors (BoC51, 109, 117, and 122) and secondary resistant tumors (BoC2 and BoC56) were harvested at the end of treatment (days 29 to 59). For BoC2 and 56, untreated control Tumors (K) were harvested once tumors reached approx. 1000mm3 in volume.

The antidepressant fluoxetine elicits anticancer effects by impairing aerobic glycolysis in colon cancer cells and xenografts.

Project description:Colon cancer is one of the most common tumors worldwide. Recent reports showed that patients treated with the antidepressant fluoxetine had reduced colon cancer risk, with effects similar to the chemotherapeutic 5-fluoro-uracil. Here, we examined the effects of fluoxetine and 5-fluoro-uracil on gene expression of HT29 colon cancer cell xenografts. HT29 xenografts in NOD/SCID mice were treated with vehicle (physiological solution), fluoxetine (30mg/kg/day), or 5-Fluoro-uracil (50 mg/kg/day).

Gene expression profiling of HhAntag-treated pancreatic xenografts

Project description:Four vehicle-treated and four HhAntag-treated pancreatic xenograft tumors were profiled for gene expression changes using Affymetrix U133 Plus 2.0 and Affymetrix Mouse Genome 430 2.0 arrays. Keywords: comparative gene expression, hedgehog, hh A primary human pancreatic tumor xenograft (1051178-A) was established by direct implantation of surgical material into female CD1 nu/nu mice of 6-8 weeks of age. Tumors were serially passaged into larger cohorts of mice for efficacy testing and subsequently distributed into tumor volume-matched cohorts upon tumors reaching between 200 to 350 mm3. HhAntag was resuspended in 0.5% methyl-cellulose, 0.2% Tween-80 (MCT) and administered orally twice daily at 75 mg/kg from a 10 mg/ml suspension. MCT alone served as vehicle control. Tumor xenografts (4/group) were excised following 21 days of dosing and RNA was extracted. Preparation of complementary RNA, Human Genome U133 Plus 2.0 array and Mouse Genome 430 2.0 array hybridizations, and subsequent data analysis were carried out using Affymetrix protocols, with signal intensities being determined by the MAS5.0 algorithm.

Gene expression changes induced by Dendrocalamus latiflorus leaf extract (DLE) and white tea extract (WTE) and pioglitazone (PGZ) in db/db mice liver

Project description:Eight-week-old C57BL/6J and T2DM db/db mice randomly divided into 4 groups of 8 based on equal bodyweight and fasting blood glucose (FBG) levels (mice were fasted for 6 h). The C57BL/6J mice (normal group) were treated with saline. The T2DM db/db mice were treated with saline, DLE (200 mg/kg), WTE (180 mg/kg), or pioglitazone (PGZ, 20 mg/kg) for 12 weeks, and were called the model group, DLE group, WTE group and PGZ group, respectively. All mice received oral administration of the indicated treatments once a day. After 12 weeks drug administration, liver aliquots from four randomly selected mice from the normal, model, PGZ and DLE groups were ground into powder in liquid nitrogen, followed by RNAseq for differentially expressed gene determination and pathway enrichment analysis.

Methods: Adult male mice were exposed to Auditory Fear Conditioning training and subsequently injected with 3.0 mg/kg corticosterone, 20 mg/kg CORT108297, 80 mg/kg CORT118335, 40 mg/kg RU486 or vehicle. Dorsal hippocampi were dissected 3 hours after injection and snap-frozen. Total RNA was isolated and send for 100bp paired-end sequencing by BGI.
