Effect of 4-HDHA during TNF-induced HT-29 cells apoptosis

ABSTRACT: Ulcerative colitis (UC) is an inflammatory disease characterized by an uncontrolled inflammatory response. Previous study showed that the immunological impairment elicted the alteration of inflammatory mediators, and ferroptosis was implicated with the lethal accumulation of reactive oxygen species (ROS).We performed lipidomics sequencing of TNF-a treated cell line mimicking the colitis in vivo.Surprisingly, we found that 4-HDHA,an autoxidation product of docosahexaenoic acid (DHA),had anti-apoptosis effect on colon epithelial cell line.To evaluate the underlying mechanism of the specific lipid perioxidin’s anti-apoptosis effect, we performed the experiment using high-throughput sequencing and get data to explain the reason why 4-HDHA could decrease TNF-induced cell apoptosis.

Effect of ferrostatin-1 and liproxstatin-1 during TNF-induced HT-29 cells apoptosis

Project description:Ulcerative colitis (UC) is an inflammatory disease characterized by an uncontrolled inflammatory response. Previous study showed that the immunological impairment elicted the alteration of inflammatory mediators, and ferroptosis was implicated with the lethal accumulation of reactive oxygen species (ROS).We have already found that lipid ROS level was elevated in both murine DSS colitis model and TNF-a-induced cell apoptosis model in vitro.Then we here use two ferroptosis inhibitors ferrostatin-1 and liproxstatin-1 trying to eliminate lipid ROS in order to relive the symptoms of colitis.But we strikingly notice that liproxstatin-1 could increase TNF-induced cell apoptosis instead of protecting cells from death as we expected before while the other inhibitor ferrostatin-1 could protect cells from death indeed.To evaluate the underlying mechanism, we performed the experiment using high-throughput sequencing and get data to explain the reason why liproxstatin-1 could increase TNF-induced cell apoptosis .

SWATH differential abundance proteomics and cellular assays show in vitro anticancer activity of arachidonic and docosahexaenoic acid monoacylglycerols in HT-29 colorectal cancer cell line

Project description:Colorectal cancer (CRC) is one of the most common and mortal types of cancer. There is increasing evidence that some polyunsaturated fatty acids (PUFA) exercise specific inhibitory actions on cancer cells through different mechanisms, as a previous study on the effect on CRC cells of two PUFA free fatty acids (FFA), docosahexaenoic acid (DHA, 22:6n3) and arachidonic acid (ARA, 20:4n6)-FFA, shown. Here we have used the same study design and technology to investigate the actions of DHA and ARA-monoacylglycerols (MAG), and we have compared the results with the previous study of the corresponding FFA. Cell assays revealed that ARA- and DHA-MAG exercised dose- and time-dependent antiproliferative actions, with DHA-MAG acting on cancer cells more efficiently than ARA-MAG. SWATH-MS massive quantitative proteomics, validated by parallel reaction monitoring and followed by pathway analysis, revealed that DHA-MAG had a massive effect in the proteasome complex, while ARA-MAG main effect was related to DNA replication. Prostaglandin synthesis also resulted inhibited by DHA-MAG. Results clearly demonstrated the ability of MAGs to induce cell death in colon cancer cells and suggested a direct relationship between chemical structure and effect.

Transcriptional profiling of HT-29 cells with different treatments

Project description:Through RNA-seq, we investigated the transcriptional changes of necroptotic cell induced by TNF-α, SM-164 and zVAD. We report here that necroptosis leads to transcriptional activation of a large set of genes including pro-inflammatory cytokines, which is much stronger than that triggered by TNF-α alone. Furthermore, the robust induction of cytokine expression could be blocked by necroptotic inhibitor Nec-1s. Thus, we show that necroptosis exerts a profound effect on transcription, and activate the expression of pro-inflammatory genes, providing insights into the mechanism by which necroptosis promotes inflammation in human diseases.

Transcription profiling of blood cells from hypertriglyceridemic men treated with docosahexaenoic acid shows suppression of LDLR and OLR1 gene expression.

Project description:Previously published results from our double-blind, placebo-controlled parallel study with docosahexaenoic acid (DHA) supplementation (3 g/d, 90 d) to hypertriglyceridemic men (39-66yr) showed that DHA reduced several risk factors for cardiovascular disease (CVD), including the plasma concentration of inflammatory markers. To determine the effect of DHA supplementation on the global gene expression pattern, we performed Affymetrix GeneChip microarray analysis of blood cells (treated with lipopolysaccharide (LPS) or vehicle) drawn before and after the supplementation from the hyperlipidemic men who participated in the previous study. Genes that were significantly differentially regulated by the LPS treatment and DHA supplementation were identified. Differential regulation of 18 genes was then confirmed by quantitative RT-PCR. Both microarray and qRT-PCR data showed that the expression of LDL receptor (LDLR), oxidized LDL receptor (OLR1), and cathepsin L1 (CTSL) was significantly suppressed by DHA supplementation; however, LPS stimulated the expression of LDLR and CTSL but not that of OLR1. LPS up-regulated and DHA suppressed the expression of prostaglandin E synthase (PTGES), PPAR delta, and various chemokines. Enrichment with Gene Ontology categories demonstrated that the genes related to transcription factor activity, immune responses, host defense responses, inflammatory responses, and apoptosis were inversely regulated by LPS and DHA. These results provide supporting evidence for the anti-inflammatory effects of DHA supplementation, and reveal previously unrecognized genes that are regulated by DHA, and are associated with risk factors of cardiovascular diseases. Lactobacilli secretome-mediated negative regulator induction in HT-29 human IEC

Project description:Recent evidence suggests that lactic acid bacteria (LAB) communicate with host cells via secretome components to influence immune responses, but less is known about gut pathogen secretomes, impact of LAB secretomes on host-pathogen interactions and the mechanisms underlying these interactions. Genome-wide microarrays and cytokine profiling were used to interrogate the impact of the Lactobacillus rhamnosus R0011 secretome (LrS) on TNF-α and Salmonella enterica subsp. enterica serovar Typhimurium secretome (STS)-induced outcomes in human intestinal epithelial cells (IECs). The LrS attenuated both TNF-α- and STS-induced gene expression involved in NF-κB and MAPK activation, as well as expression of genes involved in other immune-related signaling pathways. Specifically, the LrS induced the expression of dual specificity phosphatase 1 (DUSP1), activating transcription factor 3 (ATF3) and tribbles pseudokinase 3 (TRIB3), negative regulators of innate immune signaling, in HT-29 IECs challenged with TNF-α or STS. TNF-α and STS-induced acetylation of H3 and H4 histones was attenuated by the LrS, as was the production of TNF-α and STS-induced pro-inflammatory cytokines and chemokines. Interestingly, the LrS induced production of macrophage migration inhibitory factor (MIF), a cytokine involved in host-microbe interactions at the gut interface. We propose that the LrS attenuates pro-inflammatory mediator expression through increased transcription of negative regulators of innate immune activity and changes in global H3 and H4 histone acetylation. These findings provide novel insights into the complex multifaceted mechanisms of action behind secretome-mediated interdomain communication at the gut-mucosal interface.

Docosahexaenoic acid attenuates metabolic dysfunctions induced by lipopolysaccharide in BV-2 microglial cells

Project description:Microglial cell activation has been linked to many neurodegenerative diseases. Upon stimulation by lipopolysaccharide (LPS), a number of proteins involved in inflammatory and oxidative pathways are activated. Production of nitric oxide has been regarded as a signature marker of inflammatory responses. Our recent studies demonstrated the effects of docosahexaenoic acid (DHA) to inhibit the LPS-induced inflammatory responses in BV-2 microglial cells. DHA also can upregulate the anti-oxidative pathway involving nuclear factor erythroid 2-Like 2 (Nrf2) and synthesis of heme oxygenase-1 (HO-1), a potent anti-oxidative enzyme. In order to further understand the proteins involved, this study used a label-free quantitative proteomics approach to examine effects of DHA and LPS on proteins and signaling pathways in microglial cells.

EFHD2 Suppresses Intestinal Inflammation by Blocking Intestinal Epithelial Cell TNFR1 Internalization and Cell Death

Project description:Death of intestinal epithelial cells (IECs) is important in the pathological process of intestinal inflammatory diseases. TNF acts as one pathogenic driver for inducing IEC death and substantial intestinal inflammation. However, the physiological protective mechanisms for suppressing TNF-induced IEC death remain poorly understood. Here, we report that EF-hand domain-containing protein D2 (EFHD2), highly expressed in normal intestine tissues but decreased in intestinal biopsy samples of ulcerative colitis patients, protects the intestinal epithelium from TNF-induced IEC apoptosis. EFHD2 inhibits TNF-induced apoptosis in primary IECs and intestinal organoids (enteroids). Mice deficient of Efhd2 in IECs exhibit excessive IEC death and exacerbated experimental colitis. Mechanistically, EFHD2 interacts with Cofilin to suppress the phosphorylation of Cofilin, leading to the inhibition of TNF receptor Ⅰ (TNFR1) internalization and TNF-induced apoptosis. Our findings define EFHD2 as an endogenous suppressor of IEC death to protect intestine inflammation, providing new insight to the regulation of death receptor signaling and control of intestinal inflammatory diseases.

mRNA expression profiles (transcriptome) of human endothelial cells (cell line: TIME)

Project description:We report the transcriptome of human endothelial cells (cell line: TIME) after supplementation with polyunsaturated fatty acids (PUFA) and/or stimulation with pro-inflammatory cytokines. The fatty acids docosahexaenoic acid (DHA, C22:6n3) or arachidonic acid (AA, C20:4n6) were included in the culture medium in concentrations of 15 µmol/L using ethanol as a vehicle (0.2 % v/v final ethanol concentration). Cells were cultured in the enriched media totaling 144 h. Stimulation of cells was performed in the last 24 h of fatty acid supplementation by addition of the cytokines IL-1β, TNF-α, and IFN-γ each in a concentration of 5 ng/ml.
