Project description:Experiment for label-free quantification of contaminants in preparations of EXTL3ΔN from transgenic HEK cells. EXTL3ΔN-expressing HEK cells were not untreated (untransfected / Batch C), treated with a set of CRISPR control plasmids (CRISPR control / Batch A), or treated with a set of CRISPR plasmids targeting EXT1 (CRISPR EXT1 / Batch B). EXTL3ΔN was then purified by HisTrap and size exclusion before analysing by mass spectrometry.

Project description:We identified a new RhoE isoform, RhoEα, and compared the differentially regulated signalings by these two proteins. Both RhoE and RhoEα were first knocked out via CRISPR/Cas9 in HEK 293 cells and HeLa cells (KO cells), followed by RhoE and RhoEα reintroduction (RhoE reintroduction cells and RhoEα reintroduction cells, resepectively). Wild-type cells, KO cells, RhoE reintrocution cells, and RhoEα reintroduction cells were then subjected to RNA-seq analysis.

Project description:We generated 6 cell lines disrupting the MIA3 gene using CRISPR/Cas9. We have then run RNAseq to compare these 6 lines to the control (Wild-type (WT) parental cell line). The aim of the study was to define changes in the transcriptome following loss of different isoforms of the TANGO1 protein encoded by the Mia3 gene. These data are linked to proteomic data submitted to PRIDE with accession numbers PXD024214 and PXD024221.
