The target gene investigation in osteoblats (OBs) treated with mmu-miR-1963 enriched in EVs from OCs with prostate cancer (PCa) cells [target_EV-miR-1963]

ABSTRACT: Bone metastatic lesions of PCa contain areas of bone destruction and formation that are directed by OCs and osteoblasts (OBs), respectively. OC-derived EVs are reported to maintain normal bone homeostasis; however, there is no study investigating the role of EVs from OCs in the presence of PCa. We identified mmu-miR-1963 enriched in EVs from OCs co-cultured with several PCa cell lines. NGS analyses were performed to investigate predicted target genes of EV-miR-1963 in OBs.

Gene expression analysis in the zebrafish fractured scale

Project description:Bone is a connective tissue which undergoes continuous remodeling, including bone resorption and formation by osteoclasts (OCs) and osteoblasts (OBs), respectively. Recently, extracellular vesicles (EVs) are increasingly appreciated as a regulator of intercellular communication between OCs and OBs. The zebrafish scale is a thin membranous bone embedded in the skin and consists of OBs, OCs, and bone matrix, providing an elegant model to visualize OC-OB communication. Here, we developed a double-transgenic zebrafish line, trap:GFP; osterix:mCherry, which expresses GFP and mCherry under the control of the OC-specific trap (tartrate-resistant acid phosphatase) and OB-specific osterix enhancer, respectively. Utilizing this double-transgenic line, in combination with Hoechst 33342 (Hoe) staining, we isolated four distinct fractions from the scale at 1 day post-fracture by flow cytometry, GFP(–) mCh(+) Hoe(high) (OB fraction), GFP(low) mCh(+) Hoe(high) (OC precursor (pOC) fraction), GFP(high) Hoe(high) (mature OC (mOC) fraction), and mCh(+) Hoe(–) (OB-derived extracellular vesicle (EV) fraction). In this study, we have performed RNA-seq analysis using these four fractions to examine the gene expression profile of each fraction.

miR-26a regulate the extracellular vesicle secretion from prostate cancer cells via targeting PFDN4, CHORDC1 and SHC4

Project description:Extracellular vesicles (EVs) are known to be involved in inter-cellular communication during cancer progression, however, the biogenesis of EVs in cancer cells is not completely unveiled. It has been shown that microRNAs (miRNAs) regulated variety of physiological and pathological phenomena, thus, miRNAs could regulate the EV secretion. Here, we have performed high throughput miRNA-based screening to identify the regulators of EV secretion using ExoScreen assay. By this miRNA-based screening, we identified miR-26a, which was reported as tumor suppressive miRNA, was found to be an miRNA involved in EV secretion from prostate cancer (PCa) cells. To study the effect of miR-26a on gene expression in EV secretion, transcriptome analysis for miR-26a overexpressing PCa cell lines was performed.

The gene expression profiles of human multiple myeloma cell lines transfected with miR-10a mimic

Project description:In multiple myeloma (MM), abnormal plasma cells interact with bone marrow (BM) stromal cells and vascular cells among others. Bone marrow stromal cells (BMSCs) interact with MM cells in the bone marrow (BM), and also create a permissive microenvironment for MM cell growth and survival. Recent evidence indicated that extracellular vesicles (EVs)-mediated MM cell-BMSC communication plays an important role in the MM microenvironment. In this study, we investigated the biological property of the EVs and EV-miRNAs derived from BMSCs, aiming to establish the emerging strategies to target MM microenvironment to prevent tumor growth and spread. We found that the MM-BMSC-EVs enhanced the cell proliferation of RPMI8226. The EV-miRNA expression was different between MM-BMSCs and Normal-BMSCs, and some miRNAs, including miR-10a, were significantly up-regulated in the MM-BMSC-EVs. We then visualized with an in vitro model the uptake of Cy3-labeled miR-10a into RPMI8226 via EVs. To identify the function of miR-10a in MM cells, miR-10a mimic was transfected into RPMI8226 cells.

mmu-miR-709 gene targets

Project description:We analyzed miRNA profiles in normal liver and identified miR-709, a highly abundant miRNA in this tissue. Through microarray analysis, gene targets were identified. Thirty-six genes were significantly down-regulated >2-fold in the miR-709-treated relative to the control group, and at least three of them, including Ras-related protein 11b (Rab11b), phosphatidylcholine transfer protein (Pctp), and carboxylesterase 1g (Ces1g), are direct targets. miR-709 regulates genes implicated in cytoskeleton organization, extracellular matrix attachment, cell proliferation and fatty acid metabolism, suggesting a coordinated response during cell division, when cytoskeleton remodeling requires substantial changes in gene expression.

Deep sequencing identified serum extracellular vesicle delivered miR-181a-5p as an indicator for bone-metastatic prostate cancer (microarray)

Project description:Purpose: The goals of this study are to compare the serum extracellular vesicle (EV) delivered miRNA levels of patients with bone-metastatic prostate cancer (PCa), non-bone -metastatic PCa and benign prostatic hyperplasia (BPH), and to identify EV-delivered microRNAs in patient’s serum as indicators for bone-metastatic PCa. Methods:Serum extracellular vesicle delivered miRNA profiles of patients with bone-metastatic PCa or non-bone -metastatic PCa or BPH were generated by miRNA chip array, using Agilent-070156 Human_miRNA_V21.0_Microarray plateform. Results: Differential analysis showed the expressions of 27 EV delivered miRNAs were significantly different between serum of patients with bone-metastatic PCa and non-bone-metastatic PCa with a p value <0.05. the expressions of 5 EV delivered miRNAs were confirmed with qRT–PCR. Conclusions: Serum EV-delivered miR-181a-5p is a promising diagnostic biomarker for bone-metastatic PCa.

Transcriptome profiling of macrophages exposed to extracellular vesicles containing miR-17 family.

Project description:Purpose: The goal of this study is to generate NGS-derived transcriptome profiles (RNA-seq) from bone marrow-derived macrophages (BMDMs) exposed to extracellular vesicles (EVs) containing the miR-17 family (miR-17 and miR-93). Methods: BMDMs were isolated from mouse bone and were exposed to lung epithelial Beas2B cell-derived EVs transfected with scrambled control RNAs, miR-17 mimics or miR-93 mimics. Whole transcriptome profiles of the exposed BMDMs (n = 4 biological replicates were pooled) were then generated using the Ion Proton™ System. Results: A wide range of transcript expressions were regulated by the EV-containing miR-17 and/or miR-93 in BMDMs. AC118255.2, Phox2a, 1700066J24Rik, and Gm20412 were effectively downregulated by both MV-miR-17 and MV-miR-93. Conclusions: Our study provides potential target transcripts of EV-containing miR-17/93 in macrophages.
