LFch potential as therapeutic for multiple myeloma

ABSTRACT: Lactoferrin-derived peptide chimera, LF chimera (LFch) is a synthetic peptide that mimics the functional unit of lactoferrin with antibacterial activity. Since antimicrobial peptides (AMPs) are known to be promising candidates for cancer treatment, in this study, we explored the potential of LFch for the treatment of multiple myeloma (MM), a malignant clonal plasma cell disease of the bone marrow. LFch effectively inhibited MM1S, MM1R, and RPMI8226 multiple myeloma cell growth and induced the early and late phase of apoptosis whereas did not a normal PBMCs. Furthermore, LFch modulated the relative gene expression of survival, apoptosis, and mitochondrial dysfunction at the transcriptional levels.

Identification of sialyl Lewisa/x-modified surface proteins in Multiple Myeloma cells by LC-MS

Project description:The project aims at identifying surface proteins that are modified by the tetrasaccharide sialyl Lewisa/x and therefore serve as selectin ligands in Multiple Myeloma. To that end, we have performed an immunoprecipitation using the Heca452 antibody (which recognizes sialyl Lewisa/x-related structures) and a matched isotype control on membrane-enriched fractions that have been generated from the sialyl Lewisa/x-enriched and parental Multiple Myeloma cell line MM1S and RPMI8226. Using this approach, we have been able to identify candidate proteins that may function as selectin ligands in myeloma.

Metabolomic profiling reveals the effects of early-life lactoferrin intervention on protein synthesis, energy production and antioxidative capacity in the liver of suckling piglets

Project description:<p>The objective of this study was to reveal the effects of early life lactoferrin intervention on the liver metabolism in the suckling piglets. Sixty newborn piglets with an average initial body weight (BW) of 1.51 ± 0.05 kg were assigned to the control (CON) group and the lactoferrin (LF) group. Piglets in the LF group were orally administered LF solution (0.5 g/kg BW per day) from 1 to 7 days of age, and the piglets in the CON group were orally administrated with the same dose of physiological saline. Plasma, jejunum and liver samples were collected on day 8 and 21. The LF piglets had a decreased level of plasma urea nitrogen on day 8 and an increased level of plasma albumin on day 21. Pathway analysis of metabolomic profiles showed that LF treatment affected amino acid metabolism in the liver of piglets. Additionally, LF treatment upregulated the gene expression of proteolytic enzyme and amino acid transporters (APA, aminopeptidases A; APN, aminopeptidases N; EAAC 1, excitatory amino acid carrier 1; Pept 1, peptide transporter 1) in the jejunum, and enhanced phosphorylation levels of mTOR and p70S6K in the liver. Moreover, LF treatment upregulated the expression of β oxidation related gene (CPT 1, carnitine palmitoyl transferase-1) and affected tricarboxylic acid (TCA) cycle in the liver on day 21. Furthermore, LF piglets had a lower level of MDA and a higher level of GSH and GSH-px in the liver mitochondria. Overall, the early life LF intervention affected protein synthesis, energy production and antioxidative capacity in the liver of neonatal piglets.</p>

Transcription profiling by array of E. coli NCTC 9001 challenged with four different cationic antimicrobial peptides

Project description:Obtaining an in depth understanding of the arms races between peptides comprising the innate immune response and bacterial pathogens is of fundamental interest and will inform the development of new antibacterial therapeutics. Many cationic antimicrobial peptides (AMPs) share a range of structural and physical features that have been linked to antibacterial activity and yet they vary dramatically in their potency towards the same bacterial target. We hypothesised that a whole organism view of AMP challenge on Escherichia coli could provide a sophisticated, bacterial perspective enabling understanding of how potency is linked to mode of action. We used a 1H NMR metabolomic approach to characterise the effect on E. coli of challenge with four structurally and physically related AMPs: magainin 2, pleurocidin, buforin II and a designed peptide comprising D-amino acids only. Sub-inhibitory conditions, where these peptides nevertheless induced a bacterial response, were identified enabling electron microscopic and transcriptomic analyses. Although some common features of the bacterial response to AMP challenge could be identified, the metabolomes, morphological changes and the vast majority of the changes in gene expression were specific to each AMP. We show the antibacterial mode of action of AMPs can be accurately predicted by comparing ontological profiles generated by transcriptomic analyses. The response of E. coli to AMP challenge is highly plastic, with the bacteria capable of deploying a multifaceted response adapted to the mode of action rather than the physical properties of the AMP.

Gene expression profiles of RPMI8226 cells after knockdown of KDM3A and KLF2

Project description:Recent studies have implicated KDM3A, which catalyzes removal of H3K9 methylation, is associated with tumorigenesis. However, the biological role of KDM3A in multiple myeloma, has not been delineated. Here we identify KDM3A-KLF2-IRF4 axis dependence in multiple myeloma. We demonstrate that knockdown of KDM3A leads to apoptosis and significant growth inhibition in myeloma cells. Mechanistically, KDM3A directly regulates myeloma cell survival factor IRF4 expression through H3K9 demethylation at its promoter. We further show that KDM3A directly regulates KLF2 expression and that knockdown of KLF2 leads to growth inhibition in myeloma cells. The goal of this analysis is to identify genes whose expression changes after shRNA-mediated knockdown of KDM3A and KLF2 using the human U133 plus 2.0 Affymetrix GeneChip in myeloma cell line (RPMI8226). Two independent experiments were performed: 1. Myeloma cell line (RPMI8226) was transduced with either shRNAs targeting KDM3A (duplicate hairpins) or luciferase (control) in duplicate. The gene expression profiles of KDM3A knockdown cells were compared with that of control cells. A total of 6 RNA samples (4 KDM3A knockdown and 2 control) were analyzed. 2. Myeloma cell line (RPMI8226) was transduced with either shRNAs targeting KLF2 (duplicate hairpins) or luciferase (control) in duplicate. The gene expression profiles of KLF2 knockdown cells were compared with that of control cells. A total of 6 RNA samples (4 KLF2 knockdown and 2 control) were analyzed.
