Single-cell RNA-seq analysis for high-dose testosterone treated LuCaP 96CR PDX tumors

ABSTRACT: Our single cell RNA sequencng (scRNA-seq) analysis of high-dose testosterone (Hi-T) treated patient derived xenograft model 96CR shows RB1 positive cells displayed a robust response to Hi-T while RB1 negative cells are partiallly resistant to Hi-T.

Perioperative Testosterone Replacement Therapy in Sarcopenic Male Colorectal Cancer Patients

Project description:With increasing age and the additional impact from the bowel cancer and the chemotherapy and/ or radiotherapy it has been described that testosterone (a male hormone produced naturally in the body) levels are reduced. Testosterone has an impact on numerous body functions including the muscle mass and quality. Previous studies have identified that muscle mass is reduced as a result of ageing but also because of the deleterious effect of cancer and chemotherapy and/or radiotherapy. There is growing evidence from published studies that patients with better muscle mass and quality, do better after surgery. Mr Jenkins and his team are therefore looking at ways, the investigators can try and prevent or reduce this muscle loss and therefore improve patient outcomes. The aim of this study is to assess whether using testosterone replacement therapy in the form of a topically applied gel daily for a total of 12 weeks, is feasible and acceptable by the patients who are diagnosed with colorectal cancer and are waiting to undergo surgery. The investigators will also collect information related to the testosterone replacement therapy such as questionnaires on the quality of life, fatigue and muscle mass, and blood biomarker changes in the blood.

Project description:Androgenic modulation of the structural integrity and functional maintenance of the epididymis have been demonstrated. Androgen receptor is a typical receptor to modulate classical testosterone signaling pathway, while accumulated studies suggested number of membranous receptors or intermediated molecules also involved in testosterone pathway. In RNAseq analysis of castration mouse model, profile of testosterone responsive genes was identified. Comibining with our in-vitro studies, an novel testosterone action which incoperated with glutamine and glutamate transportation has been proposed in epididymal epithlium cells.

A pilot proteomic study reveals different protein profiles related to testosterone and gonadotropin changes in a short-term controlled healthy human cohort

Project description:Testosterone deficiency afflicts approximately 30% of men aged from 40-89 years. It is associated with type 2 diabetes, metabolic syndrome and cardiovascular disease. However, the causality of the relationship between low testosterone and metabolic diseases is unclear. We have analysed plasma samples from thirty healthy male volunteers (19-32 years of age) with pharmacologically-induced testosterone deficiency. Blood samples were collected under three conditions: normal testosterone (baseline), ‘zero’ testosterone and ‘restored’ testosterone. As a proof-of-concept, a sample pooling approach was used to obtain one pool per condition, with each pool consisting of 30 individual samples. The proteomic profiles identified significant functional differences, related to the innate immune system, the regulation of insuline-like growth factors, collagen degradation, glycolysis/gluconeogenesis process, among others.

Strength training with suppressed testosterone level

Project description:Human subjects were randomized for treatment with a GnRH-analogue, Goserelin, which suppresses endogenous testosterone or placebo for 12 weeks. Strength training was performed during the last 8 weeks. The suppression of testosterone resulted in an attenuation of the normal muscle adaptation to strength training (increased muscle mass and strength). To identify molecular signals involved in the response to testosterone levels, biopsies were obtained 4 hours after the last training session and gene expression compared with Affymetrix 3' microarrays. This timepoint should capture goserelin effect on both constitutive expression, training induced changes as well as acute exercise induced (4 hours) differences in mRNA levels.
