Differentially expressed miRNAs in colon cancer tissues vs normal colon tissues

Project description:To investigate the role of miRNAs in the progression of colon cancer, we performed comprehensive miRMA microarray analysis on RNA derived from colon cancer tissues and normal colon tissues. We identified a novel set of colon cancer-related miRNAs. Total RNA was isolated from colon cancer tissues and normal colon tissues. Six-condition experiment: H normal tiss vs. H tumor tissue, S normal tiss vs. S tumor tissue, W1 normal tiss vs. W1 tumor tissue, M normal tiss vs. M tumor tissue, W2 normal tiss vs. W2 tumor tissue, and Y normal tiss vs. Y tumor tissue. Biological replicates: 1H normal tiss, 1H tumor tissue, 1S normal tissM-oM-<M-^L1S tumor tissue, 1W1 normal tissM-oM-<M-^L1W1 tumor tissue, 1M normal tissM-oM-<M-^L1M tumor tissue, 1W2 normal tissM-oM-<M-^L1W2 tumor tissue, 1Y normal tiss and 1Y tumor tissue, independently grown and harvested. One replicate per array.

Pituitaries of basal and stressed Crhr1 wild type and knockout mice

Project description:Pituitaries of basal and stressed Crhr1-wildtype (wt) and -knockout (ko) mice (Timpl et al., 1998) As the Crh-system and the HPA-axis are known to be crucially invovled in the onset, development and maintainance of psychiatric disorders like anxiety and depression and regulate the behavioural and endocrine stress responses the further analysis of Crhr1-dependent signaling cascades is essential to understand the molecular mechanisms behind these psychiatric diseases. In this project, new candidate genes involved in Crhr1-dependent signaling cascades were dissected using metyrapone treated stressed Crhr1-wt and -ko (Timpl et al., 1998) mice using 30 min restraint stress. Changes in mRNA expression of pituitaries of basal and stressed Crhr1-wt and -ko mice were analysed 1 and 3h, respectively, after stress begin on the Sentrix Mouse-6 Expression BeadChip.

Project description:To study the field cancerization effect, we screened systemic metastasis-related CNVs from morphologically normal colon tissues adjacent to colon cancer that had systemic metastasis. 89 systemic metastasis-related CNVs, mainly consisting of migration and invasion-, morphology-, cell death and survival-related pathways, were selected. Noticeably, LIM and senescent cell antigen-like domains 2 (LIMS 2, PINCH2) showed copy number amplification and up-regulation of mRNA expression in the non-relapsed group compared to the systemic relapse group. Colon cancer cells had lower expression of PINCH2 protein and mRNA compared with normal epithelial colon cells.

The sex difference under non-stressed and chronic-stressed conditions in C57BL/6 mice

Project description:Animal models are important tools in scientific research, whereas the animals used are usually single-sex instead of mixed-sex in the experimental design. To better understand the effect of sex difference, we compared several phenotypes between male and female C57BL/6 mice, including behavioral tests, plasma corticosterone levels, adult neurogenesis, and RNA-seq. The experiments were performed under non-stressed and chronic-stressed conditions, respectively. Overall, our results showed several differences between male and female mice in sensorimotor performance while little difference was found in anxiety, depression, learning, and memory. We did not observe a significant difference in adult neurogenesis. There was a sex difference in plasma corticosterone levels under chronic stress conditions, either in 30 min after the restraint stress or after 60min of the recovery. Yet, the corticosterone levels were equivalent between the sexes under non-stressed conditions at any time point. Furthermore, the results of RNA-seq identified the differential expression genes between male and female mice under non-stressed or chronic-stressed conditions.
