MiRNA Expression Profiling and Clinical Implications in Prostate Cancer Across Various Stages

ABSTRACT: This study aims to screen and identify microRNA (miRNA) expression profiles across different stages of prostate cancer (PCa) and benign prostatic hyperplasia (BPH) using high-throughput sequencing. The study seeks to determine whether specific miRNAs show consistent differential expression across various stages of PCa, with the goal of identifying potential biomarkers relevant to disease progression.

Project description:Objective: Benign prostatic hyperplasia (BPH) is a common disease in men over 50 years old, which seriously reduces the quality of life of patients. At present, the transcriptome changes of gene expression profile in benign prostatic hyperplasia have not been explored to a great extent. Methods: In this study, single-cell RNA sequencing was performed on the isolated prostate tissue of BPH mice, and its cell types were identified and enriched. In addition, we constructed the cell communication network and sequence analysis, revealing its receptor-ligand pair and its transmission signals with other cells. Results: The results revealed the changes in 11 cell subpopulations including B cells, DC, Endothelial cells, Epithelial cells, Fibroblast & Myofibroblast, Macrophage, Mast cells, Satellite cells, Schwann cells, Smooth Muscle cells, and T Cell & NK & NKT in the microenvironment at various stages of the occurrence and development of benign prostatic hyperplasia, and updated the understanding of Epithelial cells, Fibroblast & Myofibroblast cell markers. Conclusion: Our study promotes the understanding of cellular dynamic changes and potential molecular mechanisms in the development of BPH.

Intact N-glycopeptide signatures of immunoglobulin G molecules in plasma from patients with prostate carcinoma and benign prostatic hyperplasia

Project description:Benign prostatic hyperplasia (BPH) is difficult to discriminate from prostate carcinoma (PCa) preoperatively. The non-invasive biomarkers are necessary to reduce the burden of biopsies and improve survival quality of patients. Previous research suggests that abnormal glycosylation of immunoglobulin gamma molecules (IgGs) may associated with immunological diseases and prostate diseases. Hence, characterizing intact N-glycopeptides of IgGs that correspond to N-glycan structure with specific site information enable better understanding of the molecular pathogenesis and finding out some novel signatures in preoperative discrimination of BPH from PCa. In this study, we profiled intact N-glycopeptides of purified IgGs from 51 PCa patients and 45 BPH patients by our developed N-glycoproteomic method using hydrophilic interaction liquid chromatography enrichment coupled with high resolution LC-MS/MS, and intact N-glycopeptides quantitative analysis was performed using pGlyco 2.0 and MaxQuant softwares. Our data provided plasma IgG subclass-specific and site-specific N-glycosylation quantitative information.

Deep sequencing identified serum extracellular vesicle delivered miR-181a-5p as an indicator for bone-metastatic prostate cancer

Project description:Purpose: The goals of this study are to compare the serum extracellular vesicle (EV) delivered miRNA levels of patients with bone-metastatic prostate cancer (PCa), non-bone -metastatic PCa and benign prostatic hyperplasia (BPH), and to identify EV-delivered microRNAs in patient’s serum as indicators for bone-metastatic PCa. Methods:Serum extracellular vesicle delivered miRNA profiles of patients with bone-metastatic PCa or non-bone -metastatic PCa or BPH were generated by deep sequencing, using Illumina HiSeqTM 2500 platform Results: Using an optimized data analysis method, we mapped about 17 million sequence reads per sample. Differential analysis showed the expressions of 35 EV delivered miRNAs were significantly different between serum of patients with PCa and BPH, with a p value <0.05. the expressions of 5 EV delivered miRNAs were confirmed with qRT–PCR. Conclusions: Serum EV-delivered miR-181a-5p is a promising diagnostic biomarker for bone-metastatic PCa.

Multi-omic diagnostics of prostate cancer in the presence of benign prostatic hyperplasia

Project description:There is an unmet need for improved diagnostic testing and risk prediction for cases of prostate cancer (PCa) to improve care and reduce overtreatment of indolent disease. Here we have analysed the serum proteome and lipidome of 262 study participants by liquid chromatography-mass spectrometry, including participants with a diagnosis of PCa, benign prostatic hyperplasia (BPH), or otherwise healthy volunteers, with the aim of improving biomarker specificity. Although a two-class machine learning model was able to separate PCa from controls with sensitivity of 0.82 and specificity of 0.95, adding BPH resulted in a statistically significant decline in specificity for prostate cancer to 0.76, with half of BPH cases being misclassified by the model as PCa. A small number of biomarkers differentiating between BPH and prostate cancer were identified, including proteins in MAP Kinase pathways, as well as in lipids containing oleic acid; these may offer a route to greater specificity. These results highlight, however, that whilst there are opportunities for machine learning, these will only be achieved by use of appropriate training sets that include confounding comorbidities, especially when calculating the specificity of a test.
