Project description:Microarray analysis was performed in ooplast samples to identify gene expression profiles associated with the embryo capacity to develop to blastocyst stage A fraction of the cytoplasm from in vitro matured oocytes was collected and snap frozen. The oocytes were chemically activated and presumptive zygotes were in vitro cultured in individual medium microdrops. The embryo development was recorded and oocytes classified according to the stage of embryo development. Oocyte was considered of good quality if the development proceeded to blastocyst stage, whereas it was considered of bad quality if there was a development block between 8-16 cells stage and morula formation.

Project description:Is the transcriptome of cumulus cells a good predictor of the embryo’s developmental competence? The objective of this study was to analyze the transcriptional differences between cumulus cells from oocytes exhibiting different developmental potentials following individual in vitro embryo production and transfer by RNA‐seq. Cumulus cells were allocated into three groups according to the developmental potential of their enclosed oocyte: (1) not able to develop to the blastocyst stage (Bl–), (2) able to develop to the blastocyst stage but failing to establish a pregnancy (P–), or (3) able to develop to the blastocyst stage and to establish a clinical pregnancy (P+).

Transcriptomic evaluation of bovine embryo quality on blastocysts obtained from peri-pubertal oocyte donors [8 mo vs 14 mo]

Project description:Assisted reproduction technologies (ART) and high selection pressure observed in the dairy industry are leading towards the use of younger females for reproduction purpose, reducing the interval between generations. This situation might have an impact on embryo quality, which can affect the success rate of the procedures. This study aims to document the effect of donor age on embryo quality, on transcriptomic level, in order to caracterize the effect of using young animals for reproduction purpose. 10 young Holstein cows were used 3 times each at different ages for ovarian stimulation protocols and oocyte collections (at 8, 11 and 14 months). These oocytes were then fertilized in vitro with adult bull semen, generating 3 lots of embryos per animal. Semen from the same bull was used for all females, at all times. Each animal was used as its own control for age-effect evaluation. EmbryoGENE plateform allowed whole genome assessment of gene expression patterns at the blastocyst stage. Embryos from animals at 8 vs 14 months and at 11 vs 14 months were used for microarray hybridization. Validation was done performing RT-qPCR tests on 7 candidate genes.

Transcription profiling of chicken oocyte quality

Project description:The aim of this study was to assess the impact of oocyte competence on subsequent fertility. Based on knowledge already accessible in mammals and on bioinformatics tools including the chicken genome sequence, we focused on the expression of genes involved in the processes of fertilization and of early embryo development. The study was performed using two complementary approaches: a descriptive study of standard laying hens and then a differential study performed with hens from experimental lines expressing broad variations of achieved fertility (approximately 20 per cent). A differential kinetic study is performed on INRA lines selected on the basis of their fertility potential in purpose of hopefully access gene markers of fertility performance. Experiment Overall Design: All the samples were bissected from the F1 oocyte stage , and DPF+ and DPF- samples were compared in order to find differentially expressed genes that could explain the difference in fertility occuring between the two divergently selected lines. HG+ and HG- samples were compared in order to find differentially expressed genes that could explain the difference in fertility occuring between a single line. Indeed we hypothesize that mechanisms and genes related to fertility between 2 lines or in a single one might be different.
