Project description:Stem cells from various lineages have well known for its migration tendency toward glioma and become attractive vehicles to deliver therapeutic genes to brain tumors. However, which factors and mechanisms work in these function is not yet known. To identify those factors and mechanisms, we analyzed the brain tumor-specific gene expression profile using microarray analysis To identify genes that are differentially expressed, human brain tumor samples and normal brains were collected from brain tumor patients for RNA extraction and hybridization on Affymetrix microarrays. To obtain samples, we got patients consent to collect and analysis of sample. All of three normal brains (N2, N3, N4) and brain tumor, Gliobalstoma (T2), Ganglioglioma (T3) and shwannoma(T4), are used for experiment.

Lopes Abstract: In recent years, it became clear that a better understanding of the interactions among the main elements involved in the cancer network is necessary for the treatment of cancer and the suppression of cancer growth. In this work we propose a system of coupled differential equations that model brain tumour under treatment by chemotherapy, which considers interactions among the glial cells, the glioma, the neurons, and the chemotherapeutic agents. We study the conditions for the glioma growth to be eliminated, and identify values of the parameters for which the inhibition of the glioma growth is obtained with a minimal loss of healthy cells. Transcription profiling of human glial brain tumors

Project description:Gene expression profiling in 50 glial brain tumors and 4 normal brains using 42,000-feature cDNA microarrays (from total RNA). Tumors: 50 fresh-frozen glioma specimens subjected to standard WHO classification. Specimens included astrocytic [2 juvenile pilocytic astrocytomas, 1 low-grade astrocytic glioma, 1 anaplastic astrocytomas, 31 glioblastomas (of these 2 secondary glioblastomas and 2 gliosarcomas)], oligodendroglial [5 oligodendrogliomas, 3 anaplastic oligodendrogliomas], and 6 anaplastic oligoastrocytomas tumors. One tumor had been classified as glioneuronal neoplasm. Normal brain was purchased from Stratagene. Stratagene Universal Common Reference was used as reference RNA. The results provide insights into molecular mechanisms and pathways associated with gliomagenesis.

ATV-TfR and ATV-CD98hc biodistribution in the mouse brain

Project description:We evaluated the distributions of transferrin receptor (TfR)- and CD98 heavy chain (CD98hc)-targeting antibody transport vehicles (ATVs) in mouse brain using systemically injected fluorescently tagged ATVs. Combining FACS sorting with scRNA-seq, we report the localization of ATV-TfR and ATV-CD98hc on brain endothelial cells and their unique distribution across parenchymal brain cells.

Identification of Molecular Pathways Facilitating Glioma Cell Invasion In Situ

Project description:Gliomas are mostly incurable secondary to their diffuse infiltrative nature. Thus, specific therapeutic targeting of invasive glioma cells is an attractive concept. As cells exit the tumor mass and infiltrate brain parenchyma, they closely interact with a changing micro-environmental landscape that sustains tumor cell invasion. In this study, we used a unique microarray profiling approach on a human glioma stem cell (GSC) xenograft model to explore gene expression changes in situ in invading glioma cells compared to tumor core, as well as changes in host cells residing within the infiltrated microenvironment relative to the unaffected cortex. Replicate sets of mice (n=4) were inoculated with either of two different GSC's derived from from human glioma, and each mouse had samples taken from the tumor mass, the infiltrating area and the mouse brain parenchyma, resulting in 3 samples per animal. Extracellular vesicles in young serum contribute to restoration of age-related brain transcriptomes and cognition in old mice

Project description:We have demonstrated that the circulating extracellular vesicles (EVs) are essential to the beneficial effect of young serum on the skeletal muscle regenerative cascade. Here, we show that infusions of young serum significantly improved age-associated memory deficits, and the effect was abolished after serum depletion of EVs. RNA-seq analysis of the choroid plexus demonstrated EV-mediated effects on genes involved in barrier function and trans-barrier transport. Comparing the differentially expressed genes to the recently published chronological aging clocks revealed a reversal of transcriptomic aging in the choroid plexus. The hippocampal transcriptome demonstrated a significant upregulation of the anti-aging gene Klotho following young serum treatment and an abrogated effect after EV depletion. Subsequent transcriptomic profiling of Klotho knockout and heterozygous mice showed downregulation of genes associated with transport, exocytosis, and lipid transport, while upregulated genes were associated with activated microglia. The results of our study indicate the significance of EVs as vehicles to deliver signals from the periphery to brain and the importance of Klotho in maintaining brain homeostasis.

Methylation arrays delineate the extent of 5mC and 5hmC in human brain and glioma samples.

Project description:The link between global reductions in 5-hydroxymethylcytosine levels and cancer is well-known. In this study, we have characterized the extent and the location of 5hmC loss in brain tumors. We performed genome-wide methylation oxBS and BS analyses with the Illumina Infinium HumanMethylation450 BeadChip platform in 5 brain and 9 glioma samples and we measured the extent of 5mC and 5hmC in these normal versus tumoral conditions.
