Characterization of Mycobacterium smegmatis SigF regulon

Project description:Mycobacterium smegmatis SigF is a group III sigma factor. Its ortholog in M. tuberculosis is reported to have role in regulation and function of cell wall components. In present study we have created an M. smegmatis ΔsigF mutant by allele exchange method. M. smegmatis sigF mutant shows non pigmented phenotype and is more sensitive to hydrogen peroxide generated oxidative stress. DNA microarray analysis of M. smegmatis wild type and ΔsigF mutant suggests that SigF in this species controls the expression of several energy and central intermediary metabolism genes along with regulation of carotenoid biosynthesis.

Cell wall enrichment unveils proteomic changes in the cell wall during treatment of Mycobacterium smegmatis with sub-lethal concentrations of Rifampicin

Project description:In this study, we successfully developed two gel-free approaches to specifically look at cell wall proteins in Mycobacterium smegmatis. The cell wall was subjected to differential centrifugation, differential detergent solubilisation and phase separation to yield the genuine cell wall proteome. The protein extracts were digested by filter-assisted sample preparation for LC-MS/MS analysis on a Q-Exactive mass spectrometer and identified proteins subjected to a strict bioinformatics pipeline. This resulted in the unprecedented coverage of 96 lipoproteins, 475 membrane proteins containing at least one transmembrane helix and 73 secreted proteins. We used this gel-free approach to study the changes in the cell wall proteome during exposure of M. smegmatis to sub-lethal concentration of Rifampicin in a quantitative manner. This enabled us to characterise in detail the dysregulation of transporters such as TatB and ABC transporters, virulence factors such as MCE proteins and PknG, as well as proteins involved in cell wall and lipid synthesis.

Comparison of Cell Wall Proteomics of Drug Resistant M. smegmatis with and without Sub-inhibitory Rifampicin Exposure

Project description:We previously showed sub-lethal exposure to the front-line anti-TB drug rifampicin resulted in substantial adaptive remodelling of the proteome of the model organism M. smegmatis in the drug sensitive mc2155 strain (WT). Here we investigate whether these responses are conserved in an engineered, isogenic M. smegmatis mc2155 mutant harbouring the clinically relevant S531L rifampicin resistance-conferring mutation (SL) and distinguish responses that are specific to rifampicinâ€™s anti-bacterial activity from those that are dependent on rifampicin's presence alone. We used a cell wall enrichment strategy to focus attention on the cell wall proteome and observed 253 proteins to be dysregulated in SL bacteria as compared with 716 proteins in WT. We observed that down regulation of ABC transporters and up regulation of ribosomal machinery were conserved in the SL strain, while up regulation of transcriptional machinery, and down regulation of numerous two-component systems, were not.
