Project description:Investigation of partial genome gene expression level changes in a Desulfovibrio africanus during exponential and stationary phase growth in the presence and absence of 5 ug/L Hg2+ (as HgNO3). Desulfovibrio africanus is a known mercury methylating bacteria A 3 chip study using total RNA recovered from three separate cultures of Desulfovibrio africanus with 5 ug/L Hg during exponential phase growth, three seperate cultures of Desulfovibrio africanus with 5 ug/L Hg during stationary phase growth, three cultures of Desulfovibrio africanus without Hg during exponential phase growth, and Desulfovibrio africanus without Hg during stationary phase growth. Each chip measures the expression level of 4,585 genes and intergenic regions from Desulfovibrio africanus strain Walvis Bay on a custom Nimblegen format with 75-mer probes with tiled in 4-plex format.

Phosphoproteomic analysis indicates that the B cell cytoskeleton is disrupted after exposure to mercury

Project description:Exposure to environmental mercury (Hg) currently may contribute to immune system dysfunction and autoimmune disease. In this study phosphoproteomic analysis was used to investigate the impact of Hg2+ on a B cell line. Treatments were with 0, 2, 5, 10, 20, 50 and 100 uM Hg2+. 1713 phosphoproteins and 1937 confidently localized phosphorylation sites were identified. 161 phosphoproteins responded to Hg2+ treatment (ANOVA, 10% FDR). Hg2+ at 50 and 100 uM stimulated Tyr phosphorylation on residues in the B-cell receptor complex as well as other systems. At 20 uM and below Hg2+ primarily caused hyperphosphorylation of pSer/Thr residues and affected cytoskeletal systems. Map Kinase 1 was exceptional as it was hyperphosphorylated at Tyr 185 following 20 uM and lower Hg2+ treatments. These results contribute novel insight into the mechanism for immune system disruption by Hg2+.

Comparative transcriptional analysis of Desulfovibrio alaskensis G20, a choline utilization regulator and LysX

Project description:Expression data for Desulfovibrio alaskensis strain G20 and mutants in regulator proteins grown on lactate sulfate media and then pelleted and transferred to another media when they reached stationary phase. The Choline mutant was transferred to lacte/sulfate minimal media and choline/sulfate minimal media. The LysX mutant was transferred to minimal media with lysine and rich media. G20 was transferred to minimal media, choline/sulfate minimal media, lactate/choline/sulfate minimal media, minimal media with lysine, and rich media. We aimed to confirm or expand the regulons of each of the transposon interupted regulator mutants and compare gene expression responses of the regulators in different growth conditions. 10 samples were collected: 2 regulator mutants (2 conditions each), Desulfovibrio alaskensis G20 (5 conditions), 2 replicates for G20 minimal media condition. Control sample -G20 rich media.

The effect of heavy metals such as Cd2+, Cu2+ and Hg2+ to nitrifying bacterium, Nitrosomonas europaea, was studied in pseudo-steady state batch reactor. Under incubation of Nitrosomonas europaea with 1 ?M CdCl2 for 1 hour, transcripts for 66 of 2460 genes were found at high level, yet transcripts of 50 genes were found at low level.
